The primary platform used for pyrazinamide (PZA) susceptibility testing of is the MGIT culture system (Becton Dickinson). Since false-resistant results have been associated with the use of this system, we conducted a multicenter evaluation to determine the effect of using a reduced cell density inoculum on the rate of false resistance. Two reduced inoculum densities were compared with that prescribed by the manufacturer (designated as "BD" method). The reduced inoculum methods (designated as "A" and "C") were identical to the manufacturer's protocol in all aspects with the exception of the cell density of the inoculum. Twenty genetically and phenotypically characterized isolates were tested in duplicate by ten independent laboratories using the three inoculum methods. False-resistant results declined from 21.1% using the standard "BD" method to 5.7% using the intermediate ("A") inoculum and further declined to 2.8% using the most dilute ("C") inoculum method. The percentages of the resistant results that were false-resistant declined from 55.2% for the "BD" test to 28.8% and 16.0% for the "A" and "C" tests, respectively. These results represent compelling evidence that the occurrence of false-resistant MGIT PZA susceptibility test results can be mitigated through the use of reduced inoculum densities.

Background: The aim of this article is to evaluate the effect of different portions of Platelet Rich Fibrin (PRF) membranes and liquid-PRF, prepared by two distinct protocols/centrifuges each, on the multispecies subgingival biofilm.

Materials And Methods: PRF membranes and liquid-PRF were prepared using two protocols: centrifuge 1 uses fixed acceleration while centrifuge 2, progressive acceleration. PRF samples were introduced into device concurrently with 33-species bacterial inoculum.

The Ralstonia solanacearum Species Complex (RSSC) is the most significant plant pathogen group with a wide host range. It is genetically related but displays distinct biological features, such as restrictive geography occurrence. The RSSC comprises three species: Ralstonia pseudosolanacearum (phylotype I and III), Ralstonia solanacearum (phylotype IIA and IIB), and Ralstonia syzygii (phylotype IV) (Fegan and Prior 2005).

Owing to the massive refractory lignocellulose and leachate-organic loads, the stabilization of municipal solid waste (MSW) landfill is often prolonged, resulting in environmental burdens. Herein, various assembled multifunctional microbial inoculums (MMIs) were introduced into the semi-aerobic bioreactor landfill (SABL) to investigate the bioaugmentation impacts. Compared to control (CK) and other MMIs treatments (G1-G3), LD + LT + DM inoculation (G4) significantly increased volatile solids degradation (9.

Many patients with community-acquired diabetic foot infections (DFI) receive systemic (empirical) antibiotic treatments before surgical interventions, often by the general practitioner. Sometimes, hospital clinicians prescribe them before surgery to reduce the infection inoculum and preserve soft tissue for immediate wound closure in case of residual infection after surgery. In contrast, the international guidelines (IWGDF) advocate against presurgical antibiotic use in routine situations without severe progredient soft tissue infections and/or sepsis.
