Superheated droplet detectors (SDDs) are traditionally employed in the detection of neutrons. In this work the focus is on the detection of alpha particles using C2ClF5 as the target liquid. The alpha-droplet interaction is examined via computational studies, and a geometric model developed to describe the expected detector response. Experiments with alpha-emitting uranium- and samarium-doped SDDs at temperatures of 5-12°C confirm that the event rate is related to the size of the droplets, and are in model agreement for temperatures below 8°C; above this temperature, the acoustic sensitivity is reduced by signal attenuation as a result of the increasing bubble population, for which the addition of an attenuation coefficient restores the agreement with experiment. The results suggest the viability of a SDD-based alpha spectrometer using mono-sized droplets.

Implementing a robust platform analytical procedure for measuring adeno-associated virus vector genome titer.

The vector genome (vg) titer measurement, which is used to control patient dosing and ensure control over drug product manufacturing, is essential for the development of recombinant adeno-associated virus (AAV) gene therapy products. While qPCR and droplet digital PCR technologies are commonly implemented for measuring vg titer, chromatographic techniques with UV detectors represent promising future approaches, in line with traditional biotherapeutics. Here, we introduce a novel vg titer measurement approach using size-exclusion high-performance liquid chromatography with UV detection, which achieves excellent method precision (<2% relative SD), demonstrates linearity across a range of concentrations and varied particle content, is stability indicating, and can be bridged with existing vg titer methods.

PunctaFinder: An algorithm for automated spot detection in fluorescence microscopy images.

High-throughput biosensor screening and optimization are critical for health and environmental monitoring applications to ensure rapid and accurate detection of biological and chemical targets. Traditional biosensor design and optimization methods involve labor-intensive processes, such as manual pipetting of large sample volumes, making them low throughput and inefficient for large-scale library screenings under various environmental and chemical conditions. Attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopic imaging is a method for spatially resolved analysis of materials that combines the capabilities of ATR-FTIR spectroscopy with the use of a focal plane array detector. This paper presents the methodological aspects of adapting the ATR accessory with variable single reflection angle to the FTIR spectroscopic imaging method. The use of a variable reflection angle allows the image to be studied at different sample depths.
