Ursolic acid (UA), a natural triterpene acid, is a promising anti-cancer drug due to its inhibitory effect on tumor growth and metastasis. However, clinical translation of UA is limited by its poor water solubility and low bioavailability. To overcome these problems, herein an amphiphilic self-assembly nanodrug composed of UA, lactobionic acid (LA) and low-polyamidoamine (low-PAMAM) dendrimers is developed. This near-spherical nanodrug with a uniform size (~180 nm) demonstrated to have an enhanced cytotoxicity against liver cancer SMMC7721 cells, and could attenuate the migration and adhesion of SMMC7721 cells at non-toxic concentrations by suppressing metastasis-related protein MMP-9 expression. Furthermore, in vivo study indicates that the nanodrug exhibited prolonged circulation time in blood as well as increased AUC, MRT and C, and could effectively inhibit the tumor growth in H22 mice model. Overall, the UA-based nanodrug delivery system reported in the present work represents a novel strategy for targeted tumor therapy.

Epigallocatechin gallate (EGCG), an important active component extracted from green tea, has attracted much attention due to its multiple biological activities such as antioxidant, antibacterial, anti-inflammatory, and antitumor effects. Meanwhile, metformin (Met), a classic drug for the treatment of type 2 diabetes, exhibits additional benefits such as hypoglycemic, antioxidant, anti-inflammatory, and antitumor effects. However, metformin often causes gastrointestinal reactions when used alone, affecting patients' quality of life.

Elesclomol (ES) as an efficient Cu ionophore can specifically transport Cu into mitochondria and disrupt intracellular Cu homeostasis. Extra intracellular Cu induces cuproptosis and chemodynamic therapy (CDT), which further cascades immunogenic cell death (ICD) and activates antitumor immune responses. However, the tumor immunosuppressive microenvironment (TIM) attenuates the efficiency of the immune response.

Small molecule targeted inhibitor therapies often have several drawbacks, including limited oral bioavailability, quick metabolism, toxic effects that limit dosage, and poor water solubility. This study aims to develop a nanodrug self-delivery system that does not require a carrier by utilizing the self-assembly of camptothecin (CPT) and dihydroartemisinin (DHA). CPT/DHA nanoparticles (NPs) with varying diameters can be synthesized without requiring further carrier materials or chemical modifications by changing the CPT-to-DHA ratio (10:1, 5:1, 2:1, 1:1).

The efficacy of photodynamic therapy (PDT) for malignant tumors is significantly impeded by the short diffusion distance of reactive oxygen species (ROS) and the ROS-consuming glutathione (GSH) overexpressed in tumor cells. Therefore, enhanced PDT can be achieved by the construction of biomacromolecule-based nanodrugs that can specifically target ROS-sensitive mitochondria and deplete intracellular GSH. Herein, we synthesized the dextran-based nano-assemblies by a Graft copolymerization Induced Self-Assembly (GISA) method, in which methyl acrylate and diallyl disulfide (DADS) were copolymerized from a mixed dextran/amino dextran backbone in an aqueous medium.
