The rule of unity is based on a simple absorption parameter, Π, that can accurately predict whether or not an orally administered drug will be well absorbed or poorly absorbed. The intrinsic aqueous solubility and octanol-water partition coefficient, along with the drug dose are used to calculate Π. We show that a single delineator value for Π exist that can distinguish whether a drug is likely to be well absorbed (FA ≥ 0.5) or poorly absorbed (FA < 0.5) at any specified dose. The model is shown to give 82.5% correct predictions for over 938 pharmaceuticals. The maximum well-absorbed dose (i.e. the maximum dose that will be more than 50% absorbed) calculated using this model can be utilized as a guideline for drug design and synthesis.

Concerns have been raised that glucagon-like peptide 1 receptor agonists (GLP1-RAs) may increase the risk of thyroid cancer, but evidence remains conflicting. We therefore investigated if GLP1-RA use, compared with use of dipeptidyl peptidase-4 inhibitors (DPP-4is), was associated with thyroid cancer risk in patients with type 2 diabetes. This multisite cohort study with subsequent meta-analysis included six population-based databases from Canada (Ontario), Denmark, Norway, South Korea, Sweden, and Taiwan.

Blue thermally activated delayed fluorescent emitters are promising for the next generation of organic light-emitting diodes, yet their performance still cannot meet the requirements for commercialization. Here we establish a design rule for highly efficient and stable thermally activated delayed fluorescent emitters by introducing an auxiliary acceptor that could delocalize electron distributions, enhancing molecular stability in both the negative polaron and triplet excited state, while also accelerating triplet-to-singlet up-conversion and singlet radiative processes simultaneously. Proof-of-concept thermally activated delayed fluorescent compounds, based on a multi-carbazole-benzonitrile structure, exhibit near-unity photoluminescent quantum yields, short-lived delays and improved photoluminescent and electroluminescent stabilities.

Objective: Nontyphoidal infection is one of the most common foodborne illnesses, and its oncogenic potential has been documented in animal models. The primary goal of this study was to examine whether individuals who were exposed to enteric infection are more likely to develop colorectal cancer (CRC) than the general population through the linkage of 2 statewide public health surveillance databases.
Materials And Methods: We designed a 2-stage probabilistic linkage, starting with 17,587 records of enteric salmonellosis reported to Michigan Department of Health and Human Services between 1992 and 2020.
