An easy, quick, simple and accurate spectrofluorimetric method was recognized and validated for evaluation of sorafenib (SOR) in pure form and biologically in plasma. Cremophor RH 40 (Cr RH 40) used for enhancing the fluorescence activity of SOR in phosphate buffer (pH 7). Cr RH 40 improved the native fluorescence of SOR remarkably in water. The fluorescence spectrum of SOR was observed at 405 nm after excitation at 265 nm. The linearity appeared to be in the range of 5 to 600 ng ml for pure and from 9 to 500 ng ml for plasma using the protein precipitation (ppt) method while from 10 to 500 ng ml for plasma using liquid-liquid extraction method. The precisions and the accuracy of the estimated method gave satisfactory results. The recommended method was effectively applied for determination of SOR in human plasma with high recovery values. The results of some compounds that are possibly found in plasma were studied. The proposed method was also focused on real volunteers and a drug dissolution test.

Ferguson Plot Analysis of Chaperone ClpB from Moderate Halophile.

Protein J

January 2025

Alliance Protein Laboratories, 13380 Pantera Road, San Diego, CA, 92130, USA.

The Ferguson plot is a simple method for determining the molecular weight of native proteins and their complexes. In this study, we tested the validity of the Ferguson plot based on agarose native gel electrophoresis using multimeric chaperone protein, ClpB, derived from a moderate halophile that forms a native hexamer. The Ferguson plot showed a single band with a molecular weight of 1,500 kDa, approximately twice the size of the native hexamer.

Investigation the CMP process of 6 H-SiC in HO solution with ReaxFF molecular dynamics simulation.

Sci Rep

College of Mechanical Engineering, Zhejiang University of Technology, No.18, Chaowang Road, Hangzhou, 310014, Zhejiang Province, China.

To observe the chemical mechanical polishing (CMP) process at the atomic scale, reactive force field molecular dynamics (ReaxFF-MD) was employed to simulate the polishing of 6 H-SiC under three conditions: dry, pure water, and HO solution. This study examined the reactants on the surface of 6 H-SiC during the reaction in the HO solution, along with the dissociation and adsorption processes of HO and water molecules. The mechanisms for atom removal during the CMP process were elucidated.

Due to the sulfur's atoms' propensity to form molecules and/or polymeric chains of various sizes and configuration, elemental sulfur possesses more allotropes and polymorphs than any other element at ambient conditions. This variability of the starting building blocks is partially responsible for its rich and fascinating phase diagram, with pressure and temperature changing the states of sulfur from insulating molecular rings and chains to semiconducting low- and high-density amorphous configurations to incommensurate superconducting metallic atomic phase. Here, using a fast compression technique, we demonstrate that the rapid pressurisation of liquid sulfur can effectively break the molecular ring structure, forming a glassy polymeric state of pure-chain molecules (Am-S).

Alzheimers Dement

December 2024

Department of Neurology, University of Pennsylvania, Philadelphia, PA, USA.

Background: Limbic predominant age-related TDP-43 encephalopathy (LATE) is a common co-pathology in Alzheimer's disease (AD) and is associated with advanced cognitive impairment and severe atrophy in limbic regions. In AD, various maturation stages for tau neurofibrillary tangles have been characterized and can be selectively marked by monoclonal tau antibodies, providing insight into disease progression. Indeed, AD tau pathology progresses from an early "paperclip" conformation, marked by the MC1 epitope to a C-terminally truncated form of tau, marked by MN423 epitope.

Exposure of Mycobacteriodes abscessus clones to mucin affects bacterial phenotype.

Sci Rep

January 2025

Department of Biomedical Sciences, Carlton College of Veterinary Medicine, Oregon State University, Corvallis, USA.

In the past 20 years infections caused by Mycobacterioides abscessus have become increasingly common in patients with chronic lung conditions. The microorganisms are also resistant to a number of antibiotic classes, making treatment challenging. To begin understanding how the bacterium adapts to the lung environment, pure colonies of M.
