Spinocerebellar ataxia type 5 (SCA5) is a neurodegenerative disease caused by mutations in the cytoskeletal protein β-III-spectrin. Previously, a SCA5 mutation resulting in a leucine-to-proline substitution (L253P) in the actin-binding domain (ABD) was shown to cause a 1000-fold increase in actin-binding affinity. However, the structural basis for this increase is unknown. Here, we report a 6.9 Å cryo-EM structure of F-actin complexed with the L253P ABD. This structure, along with co-sedimentation and pulsed-EPR measurements, demonstrates that high-affinity binding caused by the CH2-localized mutation is due to opening of the two CH domains. This enables CH1 to bind actin aided by an unstructured N-terminal region that becomes α-helical upon binding. This helix is required for association with actin as truncation eliminates binding. Collectively, these results shed light on the mechanism by which β-III-spectrin, and likely similar actin-binding proteins, interact with actin, and how this mechanism can be perturbed to cause disease.

This paper aims to reveal the changing characteristics of the contribution rates of different production factors in China since the reform and opening up from two dimensions: stage and space. The study used national data from 1978 to 2021 and provincial data from 2000 to 2020, combined with methods such as C-D production function and spatial econometrics for analysis. Research has found that: (1) In terms of stage characteristics, during the structural adjustment stage (1978-1998), economic growth mainly relies on capital and labor input, and the contribution rate of land factors gradually decreases.

In this work, we show how shape matters for the ordering of red blood cells (RBCs) at a water-air interface for both artificially rigidified and sphered cells as a model system for hereditary spherocytosis. We report enhanced long-range order for spherical RBCs over disk-shaped RBCs arising from the increased local ordering of spheres relative to disks. We show that rigidity has a greater effect on the radial distribution of spherical vs disk-shaped RBCs by slightly increasing the average distance between cells.

In order to do quantitative evaluations of the age-friendliness of cities, the Age-Friendly Cities and Communities Questionnaire (AFCCQ) was developed in the Netherlands. The purpose of this study was to translate and test the validity and reliability of the AFCCQ for use in the Russian Federation, and to study the views on the age-friendliness of the city of Kazan in the Republic of Tatarstan from an intergenerational perspective.

The present study investigated the involvement of human papillomavirus (HPV)16 and HPV18 in oropharyngeal malignancies in order to understand the oncogenic mechanisms, and to identify biomarkers for early detection and treatment targets. Given the rising incidence of HPV-associated cancer, particularly in India, this holds significance in elucidating the molecular basis of these diseases. Structural validation of HPV16 and 18 oncoproteins E6 and E7 was conducted using computational tools, while gene expression profiles related to oral squamous cell carcinoma (OSCC) were analyzed to assess differential expression.
