Phase I Trial of DNX-2401 for Diffuse Intrinsic Pontine Glioma Newly Diagnosed in Pediatric Patients.

Background: There are no effective treatments for diffuse intrinsic pontine gliomas (DIPGs); these tumors cannot be surgical resected, and diagnosis is based on magnetic resonance imaging. As a result, tumor tissues for molecular studies and pathologic diagnosis are infrequent. New clinical trials are investigating novel medications and therapeutic techniques in an effort to improve treatment of patients with DIPGs.

Objective: To determine the safety, tolerability, and toxicity of an oncolytic adenovirus, DNX-2401, injected into the cerebellar peduncle in pediatric subjects with DIPG and to collect tumor samples of this type of tumor.

Methods: Phase I, single-center, uncontrolled trial. A tumor biopsy will be performed through the cerebellar peduncle, and DNX-2401 will be injected immediately after the biopsy. Standard therapy consisting of radiotherapy and chemotherapy will follow in 2 to 6 wk.

Expected Outcomes: Improvement of overall survival and quality of life in patients with DIPG and collection of tumor specimens to study the molecular profiling of these tumors.

Discussion: The aims of this trial are to contribute to the sample collection of DIPG and to offer treatment during the tumor tissue biopsy using the virus. If this virus works as expected, it could kill the tumor cells with no damage to healthy tissue, functioning as a targeted therapy. It is important to note that edema has not been observed with this virus in all trials performed to date. The information obtained through this and other similar studies may be useful for developing or improving new therapies in the battle against DIPG.

Aqueous zinc-based batteries (AZBs) are gaining widespread attention owing to their intrinsic safety, relatively low electrode potential, and high theoretical capacity. Transition metal dichalcogenides (TMDs) have convenient 2D ion diffusion channels, so they have been identified as promising host materials for AZBs, but face several key challenges such as the narrow interlayer spacing and the lack of in-deep understanding energy storage mechanisms. This review presents a comprehensive summary and discussion of the intrinsic structure, charge storage mechanisms, and key fabrication strategies of TMD-based cathodes for AZBs.

Implementation of Time-Averaged Restraints with UNRES Coarse-Grained Model of Polypeptide Chains.

Time-averaged restraints from nuclear magnetic resonance (NMR) measurements have been implemented in the UNRES coarse-grained model of polypeptide chains in order to develop a tool for data-assisted modeling of the conformational ensembles of multistate proteins, intrinsically disordered proteins (IDPs) and proteins with intrinsically disordered regions (IDRs), many of which are essential in cell biology. A numerically stable variant of molecular dynamics with time-averaged restraints has been introduced, in which the total energy is conserved in sections of a trajectory in microcanonical runs, the bath temperature is maintained in canonical runs, and the time-average-restraint-force components are scaled up with the length of the memory window so that the restraints affect the simulated structures. The new approach restores the conformational ensembles used to generate ensemble-averaged distances, as demonstrated with synthetic restraints.

Robust interfaces in anodes play a crucial role in boosting sodium-ion battery (SIB) performance. However, the fragile interfaces constructed by a two-step synthesis or artificial stack are prone to be destroyed during the charging/discharging processes, which significantly reduces the lifetime of SIBs. Here, a facile construction strategy is developed to produce robust interfaces in hollow sphere-like CoSe/nitrogen-doped carbon (HS-CoSe/NC) using intrinsic Co, N, C in metal-organic framework as precursors, which enhance the electron/ion diffusion kinetics.
