This phase I study evaluates the safety, MTD, pharmacokinetics (PK), pharmacodynamics, and preliminary anticancer activity of enavatuzumab, a humanized IgG1 antibody to the TWEAK receptor, in patients with advanced solid malignancies. Patients received escalating doses of enavatuzumab given intravenously over 60 minutes every 2 weeks. Blood was obtained for PK and biomarker assessment. Three patients were enrolled per dose level in a standard 3+3 design with response assessment by RECIST version 1.0, every 8 weeks. Thirty patients were enrolled at 6 dose levels ranging from 0.1 to 1.5 mg/kg. Dose-limiting toxicities included grade 4 (G4) lipase, G3 bilirubin, and G4 amylase elevations. There was no apparent correlation of liver or pancreatic enzyme elevation with drug exposure or the presence of liver metastases. Enavatuzumab exhibited a two-compartment linear PK model. Estimated systemic clearance was 23 to 33 mL/h with an elimination half-life of 7 to 18 days. The predicted target efficacious peak and trough concentrations occurred at 1.0 mg/kg following the second dose. There were no objective responses; 4 patients had stable disease. The MTD of enavatuzumab is 1.0 mg/kg i.v. every 2 weeks. Higher doses were not tolerated due to hepatopancreatic lab abnormalities. Further evaluation of the mechanisms of the liver and pancreatic enzyme toxicities is needed before embarking on further single-agent or combination strategies. .

Unbalanced power systems cause transformers and generators to overheat, system losses to climb, and protective devices to trigger. An optimization-based control technique for distributed generators (DG) balances demand and improves power quality in three imbalanced distribution systems with 10, 13, and 37 nodes. Each system phase has its own DG.

Imagine going left versus imagine going right: whole-body motion on the lateral axis.

Sci Rep

December 2024

Creative Robotics Lab, UNSW, Sydney, 2021, Australia.

Unlike the conventional, embodied, and embrained whole-body movements in the sagittal forward and vertical axes, movements in the lateral/transversal axis cannot be unequivocally grounded, embodied, or embrained. When considering motor imagery for left and right directions, it is  assumed that participants have underdeveloped representations due to a lack of familiarity with moving along the lateral axis. In the current study, a 32 electroencephalography (EEG) system was used to identify the oscillatory neural signature linked with lateral axis motor imagery.

BAY 2413555 is a novel selective and reversible positive allosteric modulator of the type 2 muscarinic acetylcholine (M2) receptor, aimed at enhancing parasympathetic signaling and restoring cardiac autonomic balance for the treatment of heart failure (HF). This study tested the safety, tolerability and pharmacokinetics of this novel therapeutic option. REMOTE-HF was a multicenter, double-blind, randomized, placebo-controlled, phase Ib dose-titration study with two active arms.

This study presents a comprehensive investigation into the intrinsic properties of RNiP (where R = Sm, Eu) filled skutterudite, employing the full-potential linearized augmented plane wave method within density functional theory (DFT) simulations using the WIEN2k framework. Structural, phonon stability, mechanical, electronic, magnetic, transport, thermal, and optical properties are thoroughly explored to provide a holistic understanding of these materials. Initially, the structural stability of SmNiP and EuNiP is rigorously evaluated through ground-state energy calculations obtained from structural optimizations, revealing a preference for a stable ferromagnetic phase over competing antiferromagnetic and non-magnetic phases.

Optimal CO intake in metastable water film in mesoporous materials.

Department of Civil and Environmental Engineering, and Research Centre for Resources Engineering towards Carbon Neutrality, The Hong Kong Polytechnic University, Hong Kong, China.

The feasibility of carbon mineralization relies on the carbonation efficiency of CO-reactive minerals, which is largely governed by the water content and state within material mesopores. Yet, the pivotal role of confined water in regulating carbonation efficiency at the nanoscale is not well understood. Here, we show that the maximum CO intake occurs at an optimal relative humidity (RH) when capillary condensation initiates within the hydrophilic mesopores.
