Neuronal localization of prosomatostatin mRNA in the rat brain with in situ hybridization histochemistry.

Individual neurons containing prosomatostatin mRNA were identified with in situ hybridization histochemistry. Our results demonstrate a widespread distribution of prosomatostatin mRNA in several regions of the rat central nervous system. Neurons containing this transcript were most abundant in the anterior olfactory nucleus, hypothalamus, hippocampus, and amygdala as well as in all regions of the cerebral cortex. Moreover, the distribution of mRNA-containing perikarya was coextensive with the location of neurons containing somatostatin-like immunoreactivity in all areas of the brain examined. Somatostatin neurons varied in their morphology and amount of hybridization signal from region to region. The widespread distribution and regional variations in neuronal morphology and the amount of hybridization signal are consistent with a neurotransmitter and/or a neuromodulator role for somatostatin in addition to its well-established neuroendocrine role. These results demonstrate that both the peptide and its mRNA are found in perikarya in the same areas and that they are therefore the sites of synthesis for somatostatin.

Neuropeptide Y (NPY) is a neuropeptide that may be involved with emotional regulation and drug addiction and may act as a neuroprotective agent during toxic insults, such as is associated with multiple, high doses of methamphetamine (METH). The purpose of the present study was to elucidate the nature of METH-induced changes in the NPY system by examining the effect of multiple, high doses of METH on preproNPY (ppNPY) mRNA expression in the striatum and the role that dopamine (DA) D1 and D2 receptors might play in these changes. Postganglionic parasympathetic neurons in guinea-pig cardiac ganglia exhibit choline acetyltransferase (ChAT)-immunoreactivity, and a large fraction (60%) of the ChAT-positive cardiac neurons co-express somatostatin-immunoreactivity. This co-expression remained when the cardiac ganglia explants were maintained in culture for 72 h (40% somatostatin-immunoreactive). The guinea-pig cardiac ganglia neurons express the high affinity pituitary adenylate cyclase activating polypeptide (PACAP)-selective PAC1 receptor, and treatment of the ganglia explants with 20 nM PACAP27 for 72 h to evaluate PACAP regulation of somatostatin expression revealed a dramatic 85% decrease in the number of somatostatin-IR neurons (6% somatostatin-IR neurons) compared with untreated control explant preparations.

The melanin-concentrating hormone (MCH) gene encodes two proteins, pro-MCH and MCH-gene-overprinted polypeptide (MGOP), produced through alternative splicing of the primary transcript. Recently, our laboratory has identified three distinct pre-pro-somatostatin (PSS) genes in goldfish brain: PSS-I encodes for somatostatin (SRIH)-14, PSS-II encodes SRIH-28, which contains [Glu(1), Tyr(7), Gly(10)] SRIH-14 at its C-terminus, and PSS-III encodes [Pro(2)] SRIH-14. In goldfish, increasing levels of the sex steroid estradiol increase the plasma levels of growth hormone (GH). However, whether sex steroids act at the level of the brain to regulate GH release is unclear.

To understand the mechanisms whereby recurrent hypoglycemia increases the risk of subsequent hypoglycemia, it was necessary to differentiate the effects of recurrent hyperinsulinemia from those of hyperinsulinemic hypoglycemia. We examined basal and hypoglycemic endocrine function in normal rats, streptozotocin-diabetic controls, and diabetic rats exposed to 4 days of 2 episodes/day of hyperinsulinemic hypoglycemia (DH) or hyperinsulinemic hyperglycemia (DI). DH and DI rats differentiated the effects of hyperinsulinemia from those of hypoglycemia.
