In our 2016 article in Scientific Reports, we advanced a new hypothesis for the Mongol withdrawal from Hungary in 1242 CE, based on a joint analysis of climatic, environmental, and historical data. The re-evaluation now offered by Pinke et al. casts doubt on this hypothesis. However, their arguments are based on a level of generality that fails to appreciate the specific conditions of the Mongol invasion, do not offer new or different climatic data, and are supported by anachronistic production data and environmental information, which cannot be related to the period in question. While we acknowledge the importance of an open debate, we stand by our conclusions.

Background: Down syndrome (DS) is characterized by the overexpression of the amyloid precursor protein gene (APP) and pro-inflammatory genes, leading to progressive beta-amyloid (Aβ) accumulation. This accumulation in DS may exacerbate neuroinflammation, contributing to the pathogenesis of Alzheimer's Disease (AD). Experimental models suggest that aquaporin 4 (AQP4), an astrocytic water channel implicated in Aβ clearance, is mislocalized with increased Aβ burden.

The amyloid cascade hypothesis as the etiological underpinning of Alzheimer disease (AD) is supported by a large body of literature, the most influential of which are genetic studies of the 1980s and 1990s. Other evidence includes the neuropathology of Down syndrome, apparent toxicity of oligomeric amyloid-β (Aβ), interactions with apolipoprotein E, and the analogy of cardiac amyloidosis. On the other hand, there is considerable phenotypic heterogeneity among the rare familial AD kindreds, which tempers extrapolation to sporadic AD.

Brain imaging studies in complex regional pain syndrome (CRPS) have found mixed evidence for functional and structural changes in CRPS. In this cross-sectional study, we evaluated two patient cohorts from different centers and examined functional connectivity (rsFC) in 51 CRPS patients and 50 matched controls. rsFC was compared in predefined ROI pairs, but also in non-hypothesis driven analyses.
