Positional stable isotope tracer analysis reveals carbon routes during ammonia metabolism of mosquitoes.

In females, the ammonia released during blood meal digestion is partially metabolized to facilitate the disposal of excess nitrogen. In this study, we used low- and high-resolution liquid chromatography-mass spectrometry (LC/MS) techniques to investigate the role of glucose during ammonia detoxification. Mosquitoes were fed a blood meal supplemented with [1,2-C]glucose, and downstream metabolites were measured for 24 h. Quantification of [C] amino acids in the entire mosquito body was conducted without sample derivatization using selected reaction monitoring of mass transitions that are indicative of the structural position of [C] atom incorporation. Identification of unlabeled and [C] isotopologs of 43 compounds, including amino acids, amino acid derivatives, and organic acids, was performed by high-resolution LC/MS techniques. Blood-fed mosquitoes synthesized [C] metabolites in mainly 2 carbon positions from [1,2-C]glucose. [C]Ala and [C]Pro were the most abundant and rapidly labeled amino acids synthesized. Additional [C] amino acids, [C] amino acid derivatives, and [C] organic acids in 1 or 2 carbon positions were also identified. Two kinetic routes were proposed based on the incorporation of a [C] atom at position 1 in specific amino acids. Our findings provide evidence that glucose is used for ammonia detoxification and [C] uric acid synthesis through multiple metabolic pathways, uncovering a metabolic link at the carbon atomic level in ammonia metabolism of -Horvath, T. Azurin, a bacterial blue-copper protein, has garnered significant attention as a potential anticancer drug in recent years. Among twenty Pseudomonas aeruginosa isolates, we identified one isolate that demonstrated potent and remarkable azurin synthesis using the VITEK 2 system and 16S rRNA sequencing. The presence of the azurin gene was confirmed in the genomic DNA using specific oligonucleotide primers, and azurin expression was also detected in the synthesized cDNA, which revealed that the azurin expression is active.

Phthalates are synthetic compounds, well-known plasticizers, with numerous applications and reported to have adverse effects on all living organisms residing in terrestrial and aquatic environments. In this study, the rice (Oryza sativa) seedlings were exposed to di-butyl phthalate (DBP) exogenously for 7 days, with varying concentrations of 0, 200, 400, 800, and 1600 mg/L, to explore the toxicological, physiological, and biochemical consequences by measuring various parameters such as pigment, lipid, and HO (hydrogen peroxide) contents. The biochemical analysis of seedlings showed that the pigments, lipids, and HO concentrations were altered abnormally.

A cost-effective strategy is reported utilizing ionic liquid (IL), 1-hexyl-3-methylimidazolium bisulfate ([HMIM] HSO), to delaminate TiC MXene, thereby enhancing its efficiency in electrocatalyzing tryptophan (Trp) oxidation. The positively charged IL effectively intercalates within the negatively charged MXene layers, fostering structural stability through π-π stacking and electrostatic interactions. Consequently, the resulting IL-TiC composite not only maintained the inherent electronic conductivity of TiC but also significantly augmented its electrocatalytic prowess.
