The lignocellulolytic filamentous fungus Neurospora crassa is able to assimilate various mono- and oligo-saccharides. However, more than half of predicted sugar transporters in the genome are still waiting for functional elucidation. In this study, system analysis of substrate spectra of predicted sugar transporters in N. crassa was performed at genome-wide level. NCU01868 and NCU08152 have the capability of uptaking various hexose, which are named as NcHXT-1 and NcHXT-2 respectively. Their transport activities for glucose were further confirmed by fluorescence resonance energy transfer analysis. Over-expression of either NcHXT-1 or NcHXT-2 in the null-hexose-transporter yeast EBY.VW4000 restored the growth and ethanol fermentation under submerged fermentation with glucose, galactose, or mannose as the sole carbon source. NcHXT-1/-2 homologues were found in a variety of cellulolytic fungi. Functional identification of two filamentous fungal-conserved hexose transporters NcHXT-1/-2 via genome scanning would represent novel targets for ongoing efforts in engineering cellulolytic fungi and hexose fermentation in yeast.

: Diabetes is a metabolic disorder characterized by increased blood sugar levels. Early detection of diabetes could help individuals to manage and delay the progression of this disorder effectively. Machine learning (ML) methods are important in forecasting the progression and diagnosis of different medical problems with better accuracy.

Background And Aims: To explore the predictive value of the triglyceride-glucose(TyG)index combined with non-high-density lipoprotein cholesterol (Non-HDL-C) in coronary atherosclerotic heart disease (CHD).
Methods And Results: We retrospectively collected patients who were suspected of CHD and underwent coronary angiography in Yiwu Central Hospital and collected medical history, other serum biochemical evaluation and echocardiography from the enrolled population, Non-HDL-C and TyG indices were calculated, and their correlation with Gensini score was analyzed. Logistic regression analysis was used to analyze the risk factors of coronary heart disease, and ROC curves were plotted to assess the predictive value of CHD in subjects with single or multiple indices.

Microwave-assisted extraction of pectin from Dillenia indica (DI) fruit was optimized using Box-Behnken design to maximize yield and quality. Parameters such as solid:solvent (1:10-1:30), microwave power (200-600 W), and extraction time (4-10 min) were varied to determine the optimal conditions. Through experimentation, the optimized extraction parameters were identified as 1:23.

Background: The apolipoprotein E (ApoE) Ɛ4 allele is associated with a significant risk for both late-onset Alzheimer's Disease (AD) development and cerebral amyloidosis, but the degree to which cerebrospinal fluid (CSF) apoE glycosylation affects disease progression is unclear. The objective of this study was to examine the relationship of CSF apoE glycosylation with t-tau, p-tau181, and Aβ1-42 CSF levels, and to delineate the effect of the APOE4+ genotype (vs E4-) on glycosylation.
Method: Total glycosylation and apoE isoform-specific glycosylation were analyzed in baseline plasma and CSF samples from a longitudinal cohort of older individuals (n=188, ages 55 - 89) from the Alzheimer's Disease Neuroimaging Initiative (ADNI).
