is a popularly known medicinal plant extensively used in many parts of the world for treatment of various diseases. Since recent past, anaphylactic/allergic type adverse events were reported upon usage, the study aimed to evaluate the anaphylactic and anaphylactoid potential of extract and andrographolide (a major phytoactive of ). The anaphylactic potential was evaluated using active systemic anaphylaxis (ASA) assay in guinea pigs. Further, the release of allergic mediators was measured in immunoglobulin E (IgE) sensitized and non-IgE sensitized Rat Basophilic Leukemia (RBL-2H3) cell lines . extract or andrographolide sensitized guinea pigs following the challenge antigen administration orally and intravenously did not demonstrate any clinical signs of anaphylaxis. IgE sensitized and non- IgE sensitized RBL-2H3 cells treated with extract did not induce release of allergic mediators. Whereas IgE sensitized and non- IgE sensitized RBL-2H3 cells treated with andrographolide demonstrated mild to moderate release of allergic mediators. extract has no anaphylactic and anaphylactoid potential in and studies. Whereas, andrographolide effects on allergic mediators in studies needs to be scrutinized if they are of biologically important.

Formaldehyde (FA) is a ubiquitous indoor air pollutant emitted from construction, consumer, and combustion-related products, and ozone-initiated reactions with reactive organic volatiles. The derivation of an indoor air quality guideline for FA by World Health Organization in 2010 did not find convincing evidence for bronchoconstriction-related reactions as detrimental lung function. Causal relationship between FA and asthma has since been advocated in meta-analyses of selected observational studies.

IL-35 modulates Tfh2 and Tfr cell balance to alleviate allergic rhinitis.

Inflamm Res

January 2025

Department of Otolaryngology, Guangzhou Women and Children's Medical Center, Guangzhou Medical University, Guangzhou, 510623, China.

Background: Allergic rhinitis (AR) represents a persistent inflammatory condition affecting the upper respiratory tract, characterized by abnormal initiation of the immunoglobulin E (IgE)-mediated cascade. Follicular helper T (Tfh) cells and regulatory T (Tfr) cells are pivotal in orchestrating the development of IgE production in AR patients. IL-35, an anti-inflammatory cytokine, secreted by various cellular subpopulations.

Background: Midges are widely distributed globally. They can transmit numerous serious diseases as well as trigger an allergic reaction in the host. Their saliva contains a variety of proteins that act as sensitizers to stimulate the host's immune response, leading to IgE-mediated allergic symptoms.
