Macrophorins are representative examples of isoprenoid epoxycyclohexenones containing cyclized drimane moieties. We located and characterized the biosynthetic gene cluster of macrophorin from Penicillium terrestris. MacJ encoded by this cluster was characterized to be the first example of a membrane-bound type-II terpene cyclase catalyzing the cyclization of meroterpenoids via direct protonation of the terminal olefinic bond in acyclic yanuthones. The late-stage functionalization and substrate promiscuity of MacJ make it a potential biocatalyst for the synthesis of macrophorin analogues.

Introduction: Cancer cachexia (CC) is characterized by weight loss with specifically reduced skeletal muscles and adipose tissues in patients with late-stage cancer. Dihydroartemisinin (DHA), an effective antimalarial derivative of artemisinin, has been demonstrated to have anti-inflammatory and antitumor properties.

Materials And Methods: This study examined the effects of DHA on the Lewis lung carcinoma (LLC)-induced CC mouse model.

Catalytic Asymmetric Total Synthesis of (+)-Chamaecydin and (+)-Isochamaecydin and their Stereoisomers.

Angew Chem Int Ed Engl

January 2025

Jilin Province Key Lab of Green Chemistry and Process, Changchun Institute of Applied Chemistry, Chinese Academy of Sciences, 5625 Renmin Street, Changchun, Jilin, 130022, China.

A modular approach was developed for the first catalytic asymmetric total syntheses of naturally occurring C terpene quinone methides and their non-natural stereoisomers, which feature the presence of an unprecedented spiro[4.4]nonane-containing 6-6-6-5-5-3 hexacyclic skeleton. Resting on a chiral phosphinamide-catalyzed enantioselective reduction of 2,2-disubstituted cyclohexane-1,3-dione, a concise route for the synthesis of enantioenriched 6-6 bicyclic fragment was developed.

Electronic address:

Diabetic retinopathy (DR) is a blinding complication of microangiopathy. First-line therapeutic drugs are all focused on late-stage DR and have several side effects, which could not meet clinical needs. The plant-derived ginsenoside Ro (Ro) has a variety of effective anti-inflammatory, immune-regulating, and cardiovascular protective effects, but its microvascular protective effects are rarely studied.

Chemoenzymatic Synthesis of the Cyclopiane Family of Diterpenoid Natural Products.

Angew Chem Int Ed Engl

November 2024

State Key Laboratory of Natural and Biomimetic Drugs, School of Pharmaceutical Sciences, Peking University, Beijing, 100191, China.

A three-stage chemoenzymatic synthesis of the cyclopiane family and related diterpenes is reported. Deoxyconidiogenol with a 6/5/5/5-fused tetracyclic cyclopiane skeleton was first produced by an engineered E. coli host harboring the corresponding terpene cyclase PchDS.

Article Synopsis
  • - Prostate cancer is a major health issue globally, particularly in resource-limited settings like Uganda, where late-stage diagnosis contributes to high mortality rates.
  • - A study at the Uganda Cancer Institute reviewed 300 patients with metastatic prostate cancer (mPCa) over five years, finding a significant drop in overall survival from 92.4% at one year to 45.2% at five years, with factors like high Gleason scores and fewer chemotherapy cycles linked to poorer outcomes.
  • - Most patients experienced severe lower urinary tract symptoms and a high median prostate-specific antigen (PSA) level at diagnosis, indicating advanced disease, emphasizing the need for improved early detection and treatment strategies in such settings.
