Cancer stem cells (CSCs) are a rare tumorigenic population of cells found in multiple types of cancer. It has been suggested that CSCs are responsible for cancer drug resistance, metastasis and recurrence. Therefore, it is important to develop techniques to correctly sort and identify CSCs. In the current study, the sorting and identification of aldehyde dehydrogenase high (ALDH) CSCs was performed using flow cytometry. Cells from three colon cancer cell lines were cultured in serum-free medium to obtain CSCs-enriched spheroid cells. Subsequently, two subpopulations of ALDH CSCs were isolated by flow cytometry either with the use of propidium iodide (PI) or not, respectively. The two subpopulations of ALDH CSCs exhibited distinct characteristics, including stem cell related gene expression, self-renewal capacity and tumorigenicity and . Key regulators of the epithelial-mesenchymal transition (EMT), including vimentin, snail and slug were highly expressed in ALDH CSCs. Therefore, the current study indicates that PI staining prior to the sorting of ALDH CSCs by flow cytometry is an appropriate system for the study of CSCs. The current study also demonstrated that there was partial overlap between the transcriptional programs underlying the EMT and CSCs.

Cancer stem cells (CSCs) account for 0.01 to 2% of the total tumor mass; however, they play a key role in tumor progression, metastasis and resistance to current cancer therapies. The generation and maintenance of CSCs are usually linked to the epithelial-mesenchymal transition (EMT), a dynamic process involved in reprogramming cancer cells towards a more aggressive and motile phenotype with increased stemness potential.

Human aldehyde dehydrogenases (ALDHs) are a group of 19 isoforms often overexpressed in cancer stem cells (CSCs). These enzymes play critical roles in CSC protection, maintenance, cancer progression, therapeutic resistance, and poor prognosis. Thus, targeting ALDH isoforms offers potential for innovative cancer treatments.

Article Synopsis
  • Muscle invasive bladder cancer (MIBC) is a serious condition associated with high rates of metastasis and limited treatment options, where immune checkpoint inhibitors (ICIs) show potential as a new therapy.
  • A detailed analysis of tumor samples identified specific cancer stem cell-like populations, particularly those expressing IGF2BP3, which are linked to immune suppression and poorer survival outcomes in MIBC patients.
  • Research on IGF2BP3 revealed its significant role in regulating key genes and pathways, suggesting that targeting this gene could be vital for developing more effective immunotherapy strategies against MIBC.

Radiotherapy is widely regarded as the primary therapeutic modality for nasopharyngeal cancer (NPC). Studies have shown that cancer cells with high resistance to radiation, known as radioresistant cancer cells, may cause residual illness, which in turn might contribute to the occurrence of cancer recurrence and metastasis. It has been shown that cancer stem-like cells (CSCs) exhibit resistance to radiation therapy.

Triple-negative breast cancer is a heterogeneous disease with high recurrence and mortality, linked to cancer stem cells (CSCs). Our study characterized distinct cell subpopulations and signaling pathways to explore chemoresistance. We observed cellular heterogeneity among and within the cells regarding phenotyping and drug response.
