In vivo near-infrared (NIR) imaging of molecular processes at the preclinical stage promises to provide more valuable mechanistic information about pathological pathways involved in neurodegeneration. NIR imaging has the potential to improve in vivo therapeutic screening protocols by enabling noninvasive monitoring of presymptomatic responses to treatment. We have developed new NIR fluorescent contrast agents conjugated to markers of cell death, and using these agents we have identified molecular pathways associated with prion-induced neurodegeneration and determined the optimal window for meaningful therapeutic intervention in prion disease. This chapter provides a description of the synthesis and purification of our NIR cell Death (NIRD) contrast agent and the application of in vivo NIRD (iNIRD) imaging to a prion model of neurodegeneration.

Multimodal phototheranostics on the basis of single molecular species shows inexhaustible and vigorous vitality, particularly those emit fluorescence in the second near-infrared window (NIR-II), the construction of such exceptional molecules nonetheless retains formidably challenging. In view of the undiversified molecular skeletons and insufficient phototheranostic outputs of previously reported NIR-II fluorophores, herein, electron acceptor engineering based on heteroatom-inserted rigid-planar pyrazinoquinoxaline was manipulated to fabricate aggregation-induced emission (AIE)-featured NIR-II counterparts with donor-acceptor-donor (D-A-D) architecture. Systematical investigations substantiated that one of those synthesized AIE molecules, namely 4TPQ, incorporating a fused thiophene acceptor, synchronously exhibited high molar absorptivity (ε), NIR-II emission, typical AIE tendency, significant reactive oxygen species (ROS) generation, and high photothermal conversion efficiency.

Background: The treatment of transplant ureteral stricture (TUS) has been a great challenge, and there is limited experience with indocyanine green (ICG) fluorescence-guided robotic Boari flap-pelvis anastomosis to identify ureteral stenosis segments (especially long-segment) and their postoperative blood supply. We report case series of ureteral strictures treated with ICG fluorescence-guided robotic Boari flap-pelvis anastomosis in our center.
Case Description: We retrospectively collected clinical data of six patients diagnosed with long-segment even full-length TUS who underwent robotic Boari flap-pelvis anastomosis with the assistance of modified distribution of robotic ports and ICG fluorescence between June 2022 and June 2024, focusing on postoperative renal function, stenosis recurrence, and urinary fistulae.
