Background: Respiratory function is impaired in chronic obstructive pulmonary disease (COPD). Automation of multi-volume CT-based measurements of different components of breathing-related airway deformations will help understand multi-pathway impairments in respiratory mechanics in COPD.
Purpose: To develop and evaluate multi-volume chest CT-based automated measurements of breathing-related radial and longitudinal expansion of individual airways between inspiratory and expiratory lung volumes.

Introduction: Changes in the human gut microbiome have been linked to various chronic diseases, including chronic obstructive pulmonary disease (COPD). While substantial knowledge is available on the genomic features of fecal communities, little is known about the microbiome's transcriptional activity. Here, we analyzed the metatranscriptomic (MTR) abundance of MetaCyc pathways, SuperPathways, and protein domain families (PFAM) represented by the gut microbiome in a cohort of non-small cell lung cancer (NSCLC) patients with- or without COPD comorbidity.

Rationale And Objectives: Hyperpolarized Xe MRI quantifies ventilation-defect-percent (VDP), the ratio of Xe signal-void to the anatomic H MRI thoracic-cavity-volume. VDP is associated with airway inflammation and disease control and serves as a treatable trait in therapy studies. Semi-automated VDP pipelines require time-intensive observer interactions.

The prevalence of obstructive sleep apnea (OSA) is on the rise, driven by various factors including more sensitive diagnostic criteria, increased awareness, enhanced technology through at-home testing enabling easy and cost-effective diagnosis, and a growing incidence of comorbid conditions such as obesity. Treating symptomatic patients with OSA syndrome to enhance quality of life remains a cornerstone approach. However, there is a lack of consensus regarding treatment to improve cardiovascular disease (CVD) outcomes, particularly in light of overall negative results from several randomized controlled trials (RCT) indicating no benefit of positive airway pressure (PAP) therapy on primary and secondary CVD events.

Despite recent advances in chronic obstructive pulmonary disease (COPD) research, few studies have identified the potential therapeutic targets systematically by integrating multiple-omics datasets. This project aimed to develop a systems biology pipeline to identify biologically relevant genes and potential therapeutic targets that could be exploited to discover novel COPD treatments via drug repurposing or drug discovery. A computational method was implemented by integrating multi-omics COPD data from unpaired human samples of more than half a million subjects.
