Alzheimers Dement

December 2024

David Geffen School of Medicine, University of California, Los Angeles, Los Angeles, CA, USA.

Background: Progressive supranuclear palsy (PSP) is a neurodegenerative disorder involving pathological deposition of tau that includes glial inclusions and specific regional vulnerability patterns. Therapeutic developments are hampered by incomplete understanding of disease mechanisms. Few studies have examined its cell type-specific effects.

Background: Clinical studies indicate that mid-life dietary patterns are a risk factor for cognitive decline. Adherence to a Mediterranean diet (MeDi) may promote healthy brain aging in contrast to a Western diet (WD), yet these diets have not been examined in pre-clinical models. We hypothesized that consumption of the MeDi would have better cognitive performance compared to the Western diet in middle-aged rats.

Background: Mitochondria are organelles where energy production takes place via oxidative phosphorylation, thus mitochondrial function influences the organs with large energy consumption, such as the brain. Mitochondria contain their own circular genome (mtDNA), which encodes essential proteins/RNAs involved in oxidative phosphorylation. The maternal inheritance of mtDNA, combined with a higher risk of Alzheimer's disease (AD) observed in females, suggest mtDNA may have a role in AD.

Alzheimers Dement

December 2024

VA Boston Healthcare System, Boston, MA, USA.

Background: T-cell infiltration into the brain parenchyma is associated with hyperphosphorylated tau (p-tau) accumulation in neurodegenerative diseases. Chronic traumatic encephalopathy (CTE) is a progressive tauopathy caused by exposure to repetitive head impacts (RHI). CTE is defined by the perivascular accumulation of p-tau at the cortical sulcal depths and can be stratified into mild and severe pathological stages.

Alzheimers Dement

December 2024

Huashan Hospital, Fudan University, Shanghai, Shanghai, China.

Background: Cognition and its two critical proxies, socioeconomic status (SES) and educational attainment (EA), contribute substantially to human health and are heritable. Elucidating the genetic characteristics of SES/EA/Cognition not only helps to understand the innate individual differences in cognition, but also aids in unraveling the biological mechanisms of complex cognitive-related disorders such as Alzheimer's disease (AD). Here, we explored the rare and common protein-coding variants impacting the comprehensive cognition phenotypic spectrum by leveraging large-scale exomes.
