Structural Control of Kinetics for Macrocycle Threading by Fluorescent Squaraine Dye in Water.

While the general concept of steric speed bumps has been demonstrated in rotaxane shuttles and macrocycle threading systems, the sensitivity of speed bump effects has not been evaluated as a function of structural geometry. Values of K and k for macrocycle threading in water are reported for a series of homologous squaraine dyes with different substituents (speed bumps) on the flanking chains and two macrocycles with different cavity sizes. Sensitivity to a steric speed bump effect was found to depend on (a) structural location, being lowest when the speed bump was near the end of a flanking chain, and (b) macrocycle cavity size, which was enhanced when the cavity was constricted. This new insight is broadly applicable to many types of molecular threading systems.

Nanohoops, an exciting class of fluorophores with supramolecular binding abilities, have the potential to become innovative tools within biological imaging and sensing. Given the biological importance of cell membranes, incorporation of macrocyclic materials with the dual capability of fluorescence emission and supramolecular complexation would be particularly interesting. A series of different-sized nanohoops-ethylene glycol-decorated []cyclo--pyrenylenes (CPYs) ( = 4-8)-were synthesised an alternate synthetic route which implements a stannylation-based precursor, producing purer material than the previous borylation approach, enabling the growth of single-crystals of the Pt-macrocycle.

Directional Ring Translocation in a pH- and Redox-Driven Tristable [2]Rotaxane.

We describe the synthesis and characterization of a [2]rotaxane comprising a dibenzo-24-crown-8 (DB24C8) macrocyclic component and a thread containing three recognition sites: ammonium (AmH), bipyridinium (Bpy) and triazolium (Trz). AmH and Bpy are responsive to fully orthogonal stimuli, pH and electrochemical, which allows to precisely control the directional translation of the macrocycle along the axle. A better understanding of the processes driving the operation of the system was obtained thanks to an in-depth thermodynamic characterization.

Cascading Macrocycle and Helix Motions in a Foldarotaxane Molecular Shuttle.

The design of a dynamically assembled foldarotaxane was envisioned with the aim of operating as a two cascading trigger-based molecular shuttle. Under acidic conditions, both the macrocycle and helix were localized around their respective best molecular stations because they are far enough from each other not to alter the stability of complexes. The pH-dependent localization of the macrocycle along the encircled axle allowed us to modulate the association between the helical foldamer and its sites of interaction on the axle.
