Notch2 Inhibition and Kidney Cyst Growth in Autosomal Dominant Polycystic Kidney Disease.

J Am Soc Nephrol

January 2025

Department of Pharmacology, Tianjin Key Laboratory of Inflammatory Biology, Center for Cardiovascular Diseases, Haihe Laboratory of Cell Ecosystem, Key Laboratory of Immune Microenvironment and Disease (Ministry of Education), The Province and Ministry Co-sponsored Collaborative Innovation Center for Medical Epigenetics, Key Laboratory of Experimental Hematology, School of Basic Medical Sciences, Tianjin Medical University, Tianjin, China.

Background: Notch signaling, a conserved mechanism of cell-to-cell communication, plays a crucial role in regulating cellular processes such as proliferation and differentiation in a context-dependent manner. However, the specific contribution of Notch signaling to the progression of polycystic kidney disease (PKD) remains unclear.

Methods: We investigated the changes in Notch signaling activity (Notch1-4) in the kidneys of autosomal dominant PKD (ADPKD) patients and two ADPKD mouse models (early and late onset).

Mutations in the oncogene are recurrently linked to chronic lymphocytic leukemia (CLL), found in approximately 10% of CLL cases at diagnosis. Although these mutations are associated with clinical outcomes, their significance in the context of treatment with anti-CD20 monoclonal antibodies, Bruton's tyrosine kinase inhibitors, and BCL2 inhibitors remains controversial. Consequently, testing for mutations is not recommended outside of a clinical setting.

[Clinical characteristics and prognosis of children with T-lineage acute lymphoblastic leukemia: a single-center study].

Zhongguo Dang Dai Er Ke Za Zhi

December 2024

Department of Hematology, Guangzhou Women and Children's Medical Center, Guangzhou Medical University, Guangzhou 510000, China.

Objectives: To study the clinical characteristics and prognosis of T-lineage acute lymphoblastic leukemia (T-ALL) and related prognostic factors.

Methods: A retrospective analysis was conducted on the children with T-ALL who were treated with the Chinese Children's Cancer Group Acute Lymphoblastic Leukemia (CCCG-ALL) regimen in Guangzhou Women and Children's Medical Center between April 2015 and December 2022.

Results: A total of 80 children were included, with a median age of 7 years and 3 months and a male/female ratio of 6:1.

Targeting nonapoptotic cell death offers a promising strategy for overcoming apoptosis resistance in cancer. In this study, we developed Tat-Ram13, a 25-mer peptide that fuses the NOTCH1 intracellular domain fragment RAM13 with a cell-penetrating HIV-1 TAT, for the treatment of T-cell acute lymphoblastic leukemia with aberrant NOTCH1 mutation. Tat-Ram13 significantly downregulated NOTCH1-target genes in T-ALL cell lines.

Landscape and prognostic significance of oncogene drivers in metastatic castration sensitive prostate cancer.

Transl Cancer Res

November 2024

Department of Radiation Oncology, Rutgers Cancer Institute of New Jersey, Rutgers Robert Wood Johnson Medical School, Rutgers University, New Brunswick, NJ, USA.

Background: Tumor suppressors are well known drivers of cancer invasion and metastasis in metastatic castration sensitive prostate cancer (mCSPC). However, oncogenes are also known to be altered in this state, however the frequency and prognosis of these alterations are unclear. Thus, we aimed to study the spectrum of oncogene mutations in mCSPC and study the significance of these alteration on outcomes.
