New route for the synthesis of new cyanoimino- and cyanoaminopyrimidines.

A novel series of 4-aryl-2-cyanoimino-3,4-dihydro-1H-pyrimidines (aryl-CIDHPMs) was synthesized using a new method via the one-pot three-component reaction of cyanoguanidine with malononitrile and aromatic aldehydes using sodium methoxide as catalyst. These new aryl-CIDHPMs were also prepared by a classical route via reaction of cyanoguanidine with the corresponding arylidenemalononitriles under our conditions. In the same manner, the reaction of cyanoguanidine with cyclohexylidenemalononitrile and/or isatinylidenemalononitrile afforded new spiro-pyrimidines 9 and 10, respectively. A new series of polyfunctionalized 2-cyanoaminopyrimidines was obtained from the reaction of cyanoguanidine with cinnamaldehyde and/or ethoxyalkylenemalononitriles.

This study describes the development of a novel hybrid nanocatalyst that was obtained by doping magnetic g-CN with Ag nanoparticles and modifying it with arginine and cyanoguanidine (Ag@FeO-g-CN-Arg-CG). An operationally simple one-pot three-component and convenient synthesis method for a series of diverse purine analogues of 5-amino-7-(substituted)--(4-sulfamoylphenyl)-4,7-dihydro-[1,2,4]-triazolo[1,5-][1,3,5]triazine-2-carboxamide derivatives generated the reaction of 2-hydrazinyl--(4-sulfamoylphenyl)-2-thioxoacetamide, cyanoguanidine and a variety of aldehydes was achieved under green conditions. This experiment was conducted to evaluate the anti-inflammatory effect of the newly synthesized compounds using indomethacin as a reference medication; all compounds were tested for anti-inflammatory activity using the inhibition of albumin denaturation, RBC hemolysis technique and COX inhibition assay. The results showed that all evaluated compounds exhibited significant anti-inflammatory efficacy leading to excellently effective RBC membrane stabilization, inhibition of protein denaturation, and inhibition of COX enzymes when compared to those of indomethacin.

We developed and validated a bioanalytical assay to quantify delamanid and its key metabolite (DM-6705) in breast milk and aimed to quantify the secretion of these compounds in breast milk. Due to the hydrophobic nature of the analytes, special care was taken during sample preparation to prevent the formation of fatty deposits during protein precipitation. This was followed by online solid phase extraction and liquid chromatography with tandem mass spectrometry for detection.

Despite significant progress in immunotherapy and chimeric antigen receptor T cell therapy of leukemia, chemotherapy is the major treatment option for the disease. Therefore, the development of potent and safe drugs for standard and targeted chemotherapy of leukemia remains an important task for medicinal chemists. A library of 94 diverse 6-aryl-4-cycloamino-1,3,5-triazine-2-amines was prepared using a one-pot microwave-assisted protocol, which involves a three-component reaction of cyanoguanidine, aromatic aldehydes and cyclic amines, and subsequent dehydrogenative aromatization of the dihydrotriazine intermediates in the presence of alkali.

The La(OTf)-catalyzed [3+2] cycloaddition reactions for the synthesis of benzo[]oxazoles/benzofurans via quinones and 1,2-di--butyl-3-(cyanimino)diaziridine (1,3-di--butyl-2-cyanoguanidine)/vinyl azides have been explored. A series of 5-hydroxybenzofuran-4-carboxylic acid derivatives and 5-hydroxybenzo[]oxazole-4-carboxylic acid derivatives were conveniently obtained with high yields and good stereoselectivities, which could be used for further transformations to valuable compounds.
