A series of dihydropyrimidine (DHPM) derivatives bearing 1,3,4-oxadiazole moiety was designed and synthesized as monastrol analogues. The new compounds were screened for their cytotoxic activity toward 60 cancer cell lines according to NCI (USA) protocol. Seven compounds were further examined against the most sensitive cell lines, leukemia HL-60(TB) and MOLT-4. The most active compounds were 9m against HL-60(TB) (IC = 56 nM) and 9n against MOLT-4 (IC = 80 nM), more potent than monastrol (IC = 147 and 215 nM, respectively). Cell cycle analysis of HL-60(TB) cells treated with 9m and MOLT-4 cells treated with 9n showed cell cycle arrest at G2/M phase and pro-apoptotic activity as indicated by annexin V-FITC staining.

Benchtop nuclear magnetic resonance (NMR) spectroscopy has enabled the monitoring and optimization of chemical transformations while simultaneously providing kinetic, mechanistic, and structural insight into reaction pathways with quantitative precision. Moreover, benchtop NMR proton lock capabilities further allow for rapid and convenient monitoring of various organic reactions in real time, as the use of deuterated solvents is not required. The complementary role of F NMR-based kinetic monitoring in the fluorination of bioactive compounds has many benefits in the drug discovery process since fluorinated motifs additionally improve drug pharmacology.

The incorporation of the ferrocenyl moiety into a bioactive molecule may significantly alter the activity of the resulting conjugate. By applying this strategy, we designed ferrocenyl analogs of monastrol - the first low molecular weight kinesin spindle protein (KSP) inhibitor. The obtained compounds showed low micromolar antiproliferative activity towards a panel of sensitive and ABC-overexpressing cancer cells.

We herein describe a rational design of a heterogeneous catalyst composed of a dinuclear cuprate anion being immobilized electrostatically on one surface of Janus-type nanosheets while the other surface is decorated with highly hydrophobic octyl groups. The catalyst was found to be well dispersible in the organic phase of a biphasic aqueous/organic mixture. It was characterized by means of elemental analysis, atomic absorption spectroscopy, mass spectrometry, N absorption-desorption analysis, thermogravimetric analysis, scanning electron microscopy (SEM), and solid-state C and Si cross-polarization magic-angle spinning nuclear magnetic resonance spectroscopy.
