AI Article Synopsis

  • * *In a study, low-intensity ultrasound (US) significantly boosted HA production in Shiraia bambusicola, increasing mycelial HA content by 177.2% and reaching a peak total production of 247.67mg/L on day 8.
  • * *The ultrasound treatment altered cellular properties and enhanced gene expression related to HA biosynthesis and exudation, providing insights for optimizing HA production in submerged cultures.

Article Abstract

Hypocrellin A (HA), a naturally occurring fungal perylenequinone, is widely used in clinic to treat skin diseases and developed as a photodynamic therapy (PDT) agent against cancers. In this study, a low intensity ultrasound (US, 0.28W/cm at 40kHz) was conducted thrice of repeated US exposure (5-min) with an interval of 12h to stimulate HA production of Shiraia bambusicola after 72h of the initial submerged cultures. US not only increased the content of HA by 177.2% in mycelia, but stimulated the release of HA into the medium with the highest total production of HA (247.67mg/L) on day 8. US could result in the decreased pellet diameter, the enhanced membrane permeability, the alternation of membrane compounds and reactive oxygen species (ROS) generation. Furthermore, the ultrasonic treatment up-regulated the expression of some HA biosynthetic genes including polyketide synthase gene (PKS), O-methyltransferase gene (Omef), O-methyltransferase/FAD-dependent monooxygenase (Mono) and FAD/FMN-dependent oxidoreductase gene (FAD), and activated major facilitator superfamily transporter gene (MFS) for HA exudation. The enhancement of HA production was mainly due to both the stimulated cellular biosynthesis and the enhanced fungal exudation of HA. These results provide a basis for understanding the US elicitation and a valuable strategy for enhancing HA production in submerged Shiraia cultures.

In this work, magnetic molecularly imprinted polymer (MMIP) capable of selectively recognizing and adsorbing cordycepin was prepared. The MMIP was prepared using cordycepin as the template molecule, methacrylic acid and acrylamide as the functional monomer and ethylene glycol dimethacrylate as the crosslinker. The MMIP was analyzed using various techniques including transmission electron microscopy, thermogravimetric analysis, Fourier transform infrared spectroscopy, vibrating sample magnetometer and x-ray diffraction.

Species of the genus Ganoderma are important medicinal and edible mushrooms, with polysaccharides being its main active ingredient. The submerged fermentation of Ganoderma is an important source of polysaccharides in the mycelia. In this study, a single factor test and an orthogonal test were employed to optimize the medium and fermentation conditions with the objective of obtaining a high content of polysaccharide in the liquid submerged fermentation culture mycelia of Ganoderma species.

A mucus layer derived from porcine intestinal organoid air-liquid interface monolayer attenuates swine enteric coronavirus infection by antiviral activity of Muc2.

Background: The mucus layer provides the first defense that keeps the epithelium free from microorganisms. However, the effect of the small intestinal mucus layer on pathogen invasion is still poorly understood, especially for swine enteric coronavirus. To better understand virus‒mucus layer‒intestinal epithelium interactions, here, we developed a porcine intestinal organoid mucus‒monolayer model under air‒liquid interface (ALI) conditions.

Article Synopsis
  • * They altered different conditions such as carbon and nitrogen sources, salt levels, and pH, discovering that these changes significantly affected both biomass yield and the chemical diversity of the metabolites produced.
  • * The findings revealed that while adjustments altered the metabolites involved, the resulting extracts showed strong antibacterial activity against common pathogens like Staphylococcus aureus and Escherichia coli, regardless of the specific conditions used.

It is advantageous to culture the retina and other tissues at the air-liquid interface to allow for more efficient gas exchange. However, gene delivery to these cultures can be challenging. Electroporation is a fast and robust method of gene delivery, but typically requires submergence in liquid buffer for electrical current flow.
