This paper reports on the vinca alkaloid produced by a novel Nigrospora sphaerica isolated from Catharanthus roseus. Through liquid chromatography-mass spectrometry (LCMS), only the crude mycelia extract of this fungus was positive for determination of vinblastine. This vinca alkaloid was then purified by using high-performance liquid chromatography (HPLC) and tested for cytotoxicity activity using MTT assays. The breast cell line cancer (MDA-MB 231) was treated with a purified vinblastine which was intracellulary produced by N. sphaerica. The purified vinblastine from extracted leaf of C. roseus was used as a standard comparison. A positive result with a value of half maximal inhibitory concentration (IC ) of > 32 μg ml was observed compared with standard (IC ) of 350 μg ml only. It showed that a vinblastine produced by N. sphaerica has a high cytotoxicity activity even though the concentration of vinblastine produced by this endophytic fungus was only 0.868 μg ml .

Article Synopsis
  • A new virus called "Nigrospora oryzae mycovirus 1" (NoMyV1) was isolated from the fungus Nigrospora oryzae, featuring a non-segmented dsRNA genome of 2891 base pairs.
  • The virus contains two open reading frames: one encoding a protein similar to capsid proteins of unclassified viruses, and another for RNA-dependent RNA polymerase (RdRp).
  • Phylogenetic analysis shows NoMyV1 clusters with other unclassified viruses, leading to the proposal of a new genus called "Unirnavirus," distinct from the family Partitiviridae.

Six collections of ascomycetes were obtained from samples collected from dead branches and leaves of in Guizhou and Yunnan provinces, China. By incorporating multigene phylogenetic analysis (ITS, LSU, , SSU, , ) supplemented by morphological data, we establish two novel species, namely and . Molecular characterization of a novel mycotombus‑like virus isolated from the phytopathogenic fungus Nigrospora oryzae.

Arch Virol

Yunnan Academy of Tobacco Agricultural Sciences, Kunming City, 650021, Yunnan Province, P.R. China.

In this study, we identified a new mycotombus-like mycovirus from the phytopathogenic fungus Nigrospora oryzae, which was tentatively designated as "Nigrospora oryzae umbra-like virus 1" (NoULV1). The complete genome of NoULV1 is 3,381 nt long, containing two open reading frames (ORF1 and ORF2). ORF1 encodes a hypothetical protein with an unknown function, while ORF2 encodes an RNA-dependent RNA polymerase (RdRp) with a conserved RdRp domain containing a metal-binding 'GDN' triplet in motif C, which is distinct from the 'GDD' motif found in most + ssRNA mycoviruses.

Here, we describe a novel mycovirus, tentatively designated as "Nigrospora sphaerica fusarivirus 2" (NsFV2), which was isolated from the phytopathogenic fungus Nigrospora sphaerica strain HNXX-Ns20. NsFV2 has a single-stranded positive-sense RNA (+ ssRNA) genome of 6,156 nucleotides, excluding the poly(A) tail, and contains two putative open reading frames (ORFs). ORF1 encodes a large polypeptide of 1,509 amino acids (aa) containing a conserved RNA-dependent RNA polymerase (RdRp) domain and a viral helicase domain.

Indole alkaloids are privileged secondary metabolites, and their production may be achieved by the microbial biotransformation of tryptophan analogues. By feeding 1-methyl-L-tryptophan (1-MT) into the culture of endophytic GGY-3, six novel (-) and seven known indole alkaloids (-) were generated. Their structures were elucidated by means of NMR spectroscopy, experimental electronic circular dichroism (ECD) spectra, and X-ray crystallography analysis.
