Gastric cancer (GC) has one of the highest mortality rates of malignancies globally. Currently, ciRS-7, a novel circular RNA, has emerged as a potential sponge for miR-7. However, few studies on ciRS-7 in GC have been performed. In this study, we investigated the clinical significance and function of ciRS-7 in GC. First, the expression levels of ciRS-7 in 102 primary GC tissues and the matched para-carcinoma tissues were evaluated and the clinical relevance was confirmed in an independent validation cohort (n = 154). Second, the effects of ciRS-7 on miR-7, PTEN, and PI3K were evaluated. Finally, the function of ciRS-7 in GC was analyzed with cell lines and nude mice. The expression of ciRS-7 was significantly upregulated in GC tissues compared with the matched para-carcinoma tissues (P = 0.0023), and the upregulation of ciRS-7 was linked to poor survival in the testing (P = 0.0143) and validation cohort (P = 0.0061). Multivariate survival analysis revealed that ciRS-7 was probably an independent risk factor of overall survival (P < 0.05). Furthermore, overexpression of ciRS-7 blocked the miR-7-induced tumor suppression in MGC-803 and HGC-27 cells and led to a more aggressive oncogenic phenotype, via antagonizing miR-7-mediated PTEN/PI3K/AKT pathway. ciRS-7 may act as a prospective prognostic biological marker and a promising therapeutic target for GC. Circular RNA CDR1as/ciRS-7- a novel biomarker in solid tumors.

Front Oncol

November 2024

Sichuan Provincial Key Laboratory for Human Disease Gene Study, Genome Sequencing Center, Department of Laboratory Medicine, Sichuan Provincial People's Hospital, School of Medicine, University of Electronic Science and Technology of China, Chengdu, China.

Article Synopsis
  • CDR1as/ciRS-7 is identified as an oncogenic regulator, and its expression correlates with poor clinical outcomes in various solid tumors based on a meta-analysis of 17 studies including 2424 patients.
  • Higher levels of CDR1as/ciRS-7 are linked to increased tumor size, advanced TNM stage, and higher rates of lymph node and distant metastasis.
  • The findings suggest that elevated CDR1as/ciRS-7 can serve as a poor prognostic marker in cancer, indicating a need for further investigation into its role in tumor progression.

Article Synopsis
  • Parkinson's Disease (PD) is a complex neurological disorder characterized by the loss of dopamine-producing neurons, requiring new treatments beyond current symptom management.
  • Non-coding RNAs (ncRNAs) are emerging as important regulators of gene expression and cellular processes, with specific types of ncRNAs showing potential roles in dopamine signaling and PD pathology.
  • The review highlights the effects of deregulated miRNAs, the role of lncRNAs in dopaminergic neurons, and the novel circRNA ciRS-7, suggesting future therapeutic targets in ncRNAs to improve dopamine signaling and treatment for PD.

Several non-coding RNAs are known to be associated with the pathobiology and progression of multiple myeloma (MM). ciRS-7 (also known as CDR1-AS), a key oncogenic circular RNA (circRNA) that sponges miR-7-5p and other cancer-related microRNAs, was recently found to be downregulated in malignant plasma cells resistant to immunomodulatory drugs. Considering that various circRNAs have a strong potential as molecular biomarkers, we aimed to investigate the expression of ciRS-7 in plasma cell disorders, assess its prognostic importance in MM, and compare these findings with those of individuals with smoldering MM (SMM) and monoclonal gammopathy of unknown significance (MGUS).

Article Synopsis
  • Long non-coding RNAs (lncRNAs) and circular RNAs (circRNAs) are essential regulators involved in various biological processes in eukaryotes.
  • These non-coding RNAs function by influencing chromatin structure, regulating transcription, interacting with RNA-binding proteins, and acting as microRNA sponges.
  • Studying conserved lncRNAs and circRNAs across different species can help deepen our understanding of their roles and mechanisms, which is often limited when focusing on just one species.

Background: Circular RNAs (circRNAs) hold potential as diagnostic markers for colorectal cancer (CRC); however, their functional mechanisms remain incompletely elucidated. This work investigates the clinical implications of a unique set comprising six circRNAs derived from serum in CRC. Furthermore, we delve into the role of exosomal circ_0084043, originating from colorectal cancer-associated fibroblasts (CAFs), with a specific focus on its contribution to endothelial cell angiogenesis.
