The products of the one-pot assembly of acetylene and ketones in the KOH/DMSO system, 7-methylene-6,8-dioxabicyclo[3.2.1]octanes, undergo an acid-catalyzed (CFCOOH, room temperature) rearrangement to rarely substituted cyclopentadienes in good-to-excellent yields. The mechanism of the rearrangement has been supported by the isolation and corresponding transformations of two intermediates.

α-Hydroxy ketones are a crucial class of organic compounds prevalent in natural products and pharmaceutical molecules. The CO-promoted hydration of propargylic alcohols is an efficient method for the synthesis of α-hydroxy ketones. Herein, an ionic liquid (IL) was designed to catalyze this reaction individually under atmospheric CO pressure, volatile organic solvents, and additives.

Background: Lipoxygenases (LOXs) are key enzymes in the unsaturated fatty acid oxidation reaction pathway and play an important regulatory role in the synthesis of fruit aroma volatiles.

Methods: gene family members were identified in the whole genome database of bitter gourd and analyzed bioinformatically. An RT-qPCR was used to analyze the expression differences in different tissues.

Hydroalkylation of terminal alkynes is a powerful approach to the synthesis of disubstituted alkenes. However, its application is largely unexplored in the synthesis of α,β-unsaturated carbonyls, which are common among synthetic intermediates and biologically active molecules. The thermodynamically less stable -isomers of activated alkenes have been particularly challenging to access because of their propensity for isomerization and the paucity of reliable -selective hydroalkylation methods.

Article Synopsis
  • The study showcases a new method for synthesizing α-substituted propargylamides and β-acylamino ketones without using metals, leveraging nitriles, aldehydes, and terminal alkynes.
  • It utilizes BF·EtO as a Lewis acid, which provides greater control over the reaction outcomes by adjusting its concentration for better selectivity and product formation.
  • This method shows strong compatibility with various nucleophiles, allowing for the creation of multiple useful compounds, enhancing tools in organic chemistry synthesis.

Structurally constraining p-block elements has become a powerful strategy for bond activation chemistry with main group compounds. Traditionally, this approach focuses on mononuclear centers, yet applying structural constraints to systems with element-element bonds remains underexplored. In this study, we introduce a cation featuring a structural constraint-elongated P-P bond that spontaneously adds to unactivated alkynes, alkenes, aldehydes, and ketones.
