Pyrimidine containing epidermal growth factor receptor kinase inhibitors: Synthesis and biological evaluation.

Structure-based design and synthesis of pyrimidine containing reversible epidermal growth factor receptor (EGFR) inhibitors 1a-d are reported. The compounds (1a-d) inhibited the EGFR kinase activity in vitro with IC range 740 nm to 3 μm. mRNA expression of EGFR downstream target genes, that is twist, c-fos and aurora were found to be altered upon treatment with compounds 1a-d. The compounds 1a-d exhibited excellent anticancer activity at low micromolar level (3.2-9 μm) in lung, colon and breast cancer cell lines. Furthermore, compounds induced the alteration in mitochondrial membrane potential and reactive oxygen species level and. Selected compound 1b was found to increase sub-G1 population indicative of cell death, the mode of cell death was apoptotic as evident from Annexin V verses propidium iodide assay. Molecular modelling further helped to investigate the binding recognition pattern of the compounds in ATP binding EGFR domain similar to erlotinib and dissimilar to WZ4002.

The prevalence of small multi-target drugs containing a fluorinated aromatic moiety among approved drugs in the market is due to the unique properties of this halogen atom. With the aim to develop potent antidiabetic agents, a series of phenylsulfonic esters based on the conjugation of the 5-substituted 2-hydroxy-3-nitroacetophenones - with phenylsulfonyl chloride derivatives substituted with a fluorine atom or fluorine-containing (-CF or -OCF) group were prepared. Their structures were characterized using a combination of spectroscopic techniques complemented with a single-crystal X-ray diffraction (XRD) analysis on a representative example.

This contribution describes a study on the reactivities of the complexes [{κP,κC-(i-Pr)PO-Ar}Ni(μ-Br)], 1a-d (Ar: CH, a; 3-Cl-CH, b; 3-OMe-CH, c; 4-OMe-napthalenyl, d), with hydroxylamines in the presence of TEMPO˙ (TEMPO˙ = (2,2,6,6-tetramethylpiperidinyl-1-yl)oxyl). The results of this study showed that treating 1a-d with a mixture of EtNOH and TEMPO˙ did not afford the desired oxidation-induced functionalization of the Ni-aryl moiety in 1a-d, giving instead the corresponding κO-TEMPOH adducts [{κP,κC-(i-Pr)PO-Ar}Ni(Br)(κO-TEMPOH)], 3a-d (TEMPOH = -hydroxy-2,2,6,6-tetramethylpiperidine). The TEMPOH moiety in these zwitterionic compounds 3 can be displaced by a large excess of acetonitrile (MeCN), 10 equiv.

Its four new derivatives (-) and previously reported diacetylated product () have been synthesized and characterized spectroscopically followed by their screening for anti-urease potential.
