The ATM kinase is a master regulator of the DNA damage response, and can interact with more than 700 proteins in response to DNA damage. These interactions play a critical role in fine-tuning the response of ATM to multiple cellular stressors, and can play both a positive or negative role in regulating its activity. Here, we detail using protein-protein interaction methods, including co-immunoprecipitation and Glutathione-S-transferase (GST) fusion protein pull-down assays to understand the molecular interactions of ATM. These assays give valuable functional insights into the role of ATM, as they are easy to establish within the laboratory, are not overly laborious, and are easily reproducible.

The multi-step macroautophagy/autophagy process ends with the cargo-laden autophagosome fusing with the lysosome to deliver the materials to be degraded. The metazoan-specific autophagy factor EPG5 plays a crucial role in this step by enforcing fusion specificity and preventing mistargeting. How EPG5 exerts its critical function and how its deficiency leads to diverse phenotypes of the rare multi-system disorder Vici syndrome are not fully understood.

Introduction: Osteoarthritis (OA) is a chronic degenerative joint disorder characterized by an imbalance in chondrocyte metabolism. Ferroptosis has been implicated in the pathogenesis of OA. The role of Sirt1, a deacetylase, in mediating deacetylation during ferroptosis in OA chondrocytes remains underexplored.

Pathogen Biology and Immunology Research Institute, Hebei North University, Zhangjiakou, Hebei, China.

is an obligate intracellular bacterium of eukaryotic cells characterized by a unique biphasic life cycle; its biosynthesis and replication must occur within a cytoplasmic vacuole or inclusion. Certain inclusion membrane proteins have been demonstrated to mediate the interactions between intra-inclusion chlamydial organisms and the host cell. It has been demonstrated previously that the -encoded Cpn0308 localizes to the inclusion membrane; however, its function remains unknown.

Calcium/calmodulin-dependent serine protein kinase (CASK) interaction protein 1/2 (Caskin1/2) is essential neuronal synaptic scaffold protein in nervous system development. Knockouts of Caskin1/2 display severe deficits in novelty recognition and spatial memory. The tandem sterile alpha motif (SAM) domains of Caskin1/2, also conserved in their Drosophila homolog Ckn, are known to form homopolymers, yet their dynamic regulation mechanism remains unclear.

Osteoarthritis (OA) is a chronic degenerative joint disease that imposes a substantial economic burden on patients and society. The aim of this study was to investigate the effects of O-linked N-acetylglucosamine transferase (OGT) and OGT-mediated O-GlcNAcylation on cartilage injury and chondrocyte ferroptosis in OA. An OA model was established using mice for the in vivo study.
