Published: May 2017

The Ugi four-component reaction was investigated as a tool for the functionalization of peptide arrays via post-synthetic side-chain modification, mimicking post-translational processes. Additionally, as a proof of concept for the synthesis of peptidomimetics on arrays, the integration of an Ugi unit into a growing peptide chain was demonstrated.

Basic amino acid alternating copolymers exhibit exceptional antimicrobial properties and biosafety, yet their application is restricted by the complexity of the synthesis process and low molecular weight ( = 1000). In this study, we synthesized a basic amino acid alternating copolymer (Orn-Val) in only one step by the Ugi four-component condensation (Ugi'4CC), achieving high molecular weight ( = 20,000) and narrow polydispersity (PDI ≤ 1.10).

Modular Design and Scaffold-Synthesis of Multi-Functional Fluorophores for Targeted Cellular Imaging and Pyroptosis.

Angew Chem Int Ed Engl

Physical and Synthetic Biology, Faculty of Biology, Ludwig-Maximilians-Universität München, Großhadernerstr. 2-4, 82152, Planegg-Martinsried, Germany.

Fluorophores are essential tools for optical imaging and biomedical research. Their synthetic modification to incorporate new functions, however, remains a challenging task. Conventional strategies rely on linear synthesis in which a parent framework is gradually extended.

Article Synopsis
  • A four-component Ugi reaction is introduced for creating novel C-N atropisomeric peptide analogues, using common materials like anilines and aldehydes to achieve complex compounds with high yields and strong stereoselectivity.
  • Adjusting the reaction temperature allows for stereodivergent outcomes, enabling the selective formation of either diastereoisomer from the same starting materials while maintaining excellent stereocontrol.
  • Research includes detailed experimental and computational analysis of the reaction mechanism, revealing that the new atropisomeric compounds exhibit varying levels of inhibitory activity, highlighting the importance of the different atropisomers' structures in their efficacy.

Loading Lewis Acid/Base Pair on Metal Nanocluster for Catalytic Ugi Reaction.

Angew Chem Int Ed Engl

Constructing structurally robust and catalytically active metal nanoclusters for catalyzing multi-component reactions is an interesting while challenging task. Inspired by Lewis acid and Lewis base catalysis, we realized the combination of both Lewis acid and Lewis base sites on the surface of a stable gold nanocluster AuCd. The catalytic potential of AuCd in four-component Ugi reaction was explored, demonstrating high activity and exceptional recyclability.

Dynamics of epigenetic modifications such as acetylation and deacetylation of histone proteins have been shown to be crucial for the life cycle development and survival of Plasmodium falciparum, the deadliest malaria parasite. In this study, we present a novel series of peptoid-based histone deacetylase (HDAC) inhibitors incorporating nitrogen-containing bicyclic heteroaryl residues as a new generation of antiplasmodial peptoid-based HDAC inhibitors. We synthesized the HDAC inhibitors by an efficient multicomponent protocol based on the Ugi four-component reaction.
