MicroRNAs (miRNAs) play a crucial role in the regulation of many post-transcriptional processes in reproductive cells. Regulation of maternal mRNA translation and activation of zygotic mRNA are essential to successful embryonic development. Moreover, the precise development of embryonic cell and/or tissue lineages requires temporal and spatial control of gene expression, mRNA abundance, and translation into proteins, which is in part regulated via miRNA. Here, we describe some key protocols that can be utilized to detect and quantify miRNA in in vitro produced oocytes and embryos.

Objective: Clinical validation of sperm selection device ZyMōt™ for standard IVF.
Methods: The pre-clinical validation of ZyMōt™ included several steps. First, split semen preparation compared density gradient centrifugation (DGC) to ZyMōt™ with primary outcome fraction and absolute number of progressive motile sperm.

The adverse effect of ambient PM exposure on very early pregnancy (VEP) remains controversial among epidemiological studies but is supported by toxicological evidence. We adopted a multicenter retrospective cohort of 141,040 cycles to evaluate the effect of PM exposure on the VEP using the fertilization and embryo transfer platform and high-resolution PM data in China. We first investigated the association between PM exposure 1 week before and 1 week after the embryo transfer date and VEP.

Infertility is a highly prevalent disease affecting 1 in 6 couples worldwide. While there are numerous treatment options available to treat infertility, fertilization (IVF) is the most successful therapy available. IVF traditionally involves superovulation of the ovaries followed by ultrasound (US)-guided oocyte retrieval, fertilization of oocytes and culture of resultant embryos.

Purpose: We aimed to evaluate if ovarian stimulation with individualized dosing of recombinant follicle-stimulating hormone (rec-FSH) with follitropin delta compared with standard gonadotropin dosing reduce occurrence of follicular asynchrony in women with polycystic ovary syndrome (PCOS) undergoing in vitro fertilization (IVF).
Methods: Matched case-control study analyzed occurrence of follicular growth asynchrony during ovarian stimulation and IVF outcomes in women with PCOS. Follicular growth was considered to be asynchronous when one or two leading follicles were at least 4 mm larger in diameter than the rest of the cohort on day 5 and 9 of stimulation.

Purpose: This study aims to create and validate a clinical model that predict the probability of blastocyst formation in IVF/ICSI-ET cycles.
Methods: This study employed a retrospective methodology, gathering data from 4961 cleavage-stage embryos that cultured in the reproductive center's of the Fourth Hospital of Hebei Medical University between June 2020 and March 2024. 3472 were in the training set and 1489 were in the validation set when it was randomly split into the training set and validation set in a 7:3 ratio.
