Photodynamic therapy is emerging as a hopeful method for the treatment of oncological diseases. In the search of novel therapeutic strategies for colorectal cancer, in this work we reported the photocytotoxic activity of a lipophilic zinc(II) phthalocyanine on a murine colon adenocarcinoma cell line (CT26 cells). The 2,9(10),16(17),23(24) tetrakis[(2-dimethylamino)ethylsulfanyl]phthalocyaninatozinc(II), named Pc9, was encapsulated into Tetronic® 1107 polymeric poloxamine micelles (T1107) and assayed in 2D and 3D cell cultures. We showed that the formulation Pc9-T1107 was efficient to reduce cell viability after photodynamic treatment both in 2D cultures (IC 10±2nM) as well as in CT26 spheroids (IC 370±11nM). Cellular uptake of Pc9-T1107 was a time- and concentration-dependent process, being the phthalocyanine formulation mainly incorporated into lysosomal vesicles and endoplasmic reticulum cisterns, but not in mitochondria. Pc9-T1107 also induced the formation of reactive oxygen species immediately after cell irradiation. We also found that the phototoxic action of Pc9-T1107 was partially reversed in the presence of antioxidants, such as TROLOX and N-acetyl-cysteine. In addition, we showed that Pc9-T1107 treatment triggered an apoptotic cell death, as suggested by the detection of pyknotic nuclei, the reduction in the expression levels of procaspase-3 and the increase in caspase-3 enzymatic activity.

Article Synopsis
  • Sonodynamic therapy (SDT) shows potential for treating unresectable ovarian cancer due to its ability to penetrate deeper tissues and lack of phototoxicity, but its effectiveness is hindered by low targeting efficiency and the hypoxic nature of tumors.* -
  • Researchers developed a new nanoplatform called AMBC, which incorporates manganese oxide nanoparticles and a radical generator, enabling better targeting to cancer cells and improved production of reactive oxygen species when stimulated by ultrasound.* -
  • The study highlights the innovative combination of US/MR dual-mode imaging and this advanced nanoplatform to enhance treatment efficiency for ovarian cancer while overcoming challenges associated with targeting and tumor oxygen levels.*

The uptake and phototoxicity of a family of BODIPY-perylene charge transfer dyads are compared in live cancer and non-cancer cell lines to evaluate their performance in imaging and photodynamic therapy (PDT). The impact of iodination and mesylation of the meso position of the compounds on their optical properties, cell uptake and toxicity are compared. Notably, across all derivatives the probes were minimally dark toxic up to 50 μM, (the maximum concentration tested), but exhibited outstanding phototoxicity with nanomolar IC values and impressive phototoxic indices (PI, ratio of dark IC to light IC), with best performance for the mesylated iodinated derivative MB2PI, which had a PI of >218 and >8.

Photodynamic therapy (PDT) is a promising alternative treatment for localized lesions and infections, utilizing reactive oxygen species (ROS) generated by photosensitizers (PS) upon light activation. Singlet oxygen (O) is a key ROS responsible for photodynamic damage. However, the effectiveness of PS in biological systems may not correlate with the efficiency of singlet oxygen generation in homogeneous solutions.

Article Synopsis
  • The mechanisms behind the development of skin tumors are still not fully understood, particularly regarding the roles of photosensitivity and phototoxicity.
  • Recent studies have linked specific nitrosamines, contaminants in common pharmaceuticals, to gene and phototoxicity in humans, raising concerns about their potential carcinogenic effects.
  • Long-term use of contaminated medications may contribute to skin cancer risk by interfering with tumor suppressor genes and activating oncogenes, affecting overall cancer development.
