The enzymatic deglycosylation of the plant flavonoid rutin (quercetin-3-O-(6-O-α-l-rhamnopyranosyl-β-d-glucopyranoside) is usually assessed by means of high performance liquid chromatography (HPLC). We have developed a spectrophotometric method for the quantification of the released quercetin. After the enzymatic reaction, quercetin is extracted with ethyl acetate, and subsequently oxidized under basic conditions. The absorbance of quercetin autooxidation products at 320nm was correlated with the quercetin concentration by linear regression (molar extinction coefficient 23.2 (±0.3)×10Mcm). With this method, rutin-deglycosylation activity in buckwheat flour and a commercial naringinase was measured, and showed no significant differences with the results obtained by HPLC. The convenience of this method resides on the enzymatic activity quantification using the natural substrate by UV-visible spectrometry. Moreover, the simplicity and speed of analysis allows its application for a large number of samples.

The fruits and vegetables we consume as part of our diet are rich in bioactive metabolites that can prevent and ameliorate cardiometabolic diseases, cancers, and neurological conditions. Polyphenols are a major metabolite family that has been intensively investigated in this context. However, for these compounds to exert their optimal bioactivity, they rely on the enzymatic capacity of an individual's gut microbiota.

Acute lung injury (ALI) is a severe inflammatory condition characterized by excessive immune responses and oxidative stress, leading to significant tissue damage. Given the need for novel therapeutic agents, this study aimed to explore the anti-inflammatory activity and mechanisms of biotransformed root extracts (BT-PGR), which were enzymatically processed using rapidsase PL Classic from . The goal was to assess the potential of BT-PGR as a natural treatment for ALI.

Protein glycosylation is recognized as a Critical Quality Attribute for the biological and therapeutic activity of many recombinant proteins. Therefore, glycosylation should be monitored rigorously to ensure the desired quality, safety, and potency of monoclonal antibodies and other therapeutic glycoproteins. However, glycans are highly heterogeneous structures in proteins, and this poses a challenge for glycoprofile analysis.

Article Synopsis
  • The study highlights the rising incidence of inflammatory bowel disease (IBD) and investigates the impact of flavonoids from mulberry leaves, particularly their enhanced anti-inflammatory effects after enzymatic hydrolysis.
  • An enzymatic method was employed to create a hydrolysate of mulberry leaf flavonoids (EMLF), which showed superior protective effects against IBD in zebrafish compared to unhydrolyzed leaves, with mechanisms linked to purine metabolism regulation.
  • The findings indicate that enzymatic deglycosylation effectively boosts the anti-inflammatory properties of mulberry leaves, suggesting potential strategies for utilizing natural products in IBD treatment.
