Focal adhesion kinase (FAK) is an important drug target that plays a fundamental role in mediating signal transduction system. We report herein the discovery of a novel class of 1,3,4-oxadiazole-2(3H)-thione derivatives containing piperazine skeleton with improved potency toward FAK. All of the 17 new synthesized compounds were assayed for the anticancer activities against four cancer cells, HepG2, Hela, SW116 and BGC823. Because of the combination of 1,4-benzodioxan, 1,3,4-oxadiazole and piperazine ring, most of them exhibited remarkable antitumor activities. Notably, compound 5m showed the most potent biological activities (IC=5.78μM for HepG2, and IC=47.15μM for SW1116), and its anti-FAK inhibitory activity (IC=0.78μM) was also the best. Computational docking studies also showed that compound 5m has interaction with FAK key residues in the active site.

Objective: Unidentified sex differences in old-age cognition may emerge in psychometric networks, which look beyond mean scores into the unique cognitive structure of males and females. Accordingly, this study aims to examine cognition in well-functioning older males and females with psychometric network analysis.

Methods: The current cohort (N = 2,802) of community-dwelling adults (≥65 years) was derived from the Advanced Cognitive Training for Independent and Vital Elderly study.

Plasma brain-related biomarkers and potential therapeutic targets in pediatric ECMO.

Extracorporeal membrane oxygenation (ECMO) is a technique used to support severe cardiopulmonary failure. Its potential life-saving benefits are tempered by the significant risk for acute brain injury (ABI), from both primary pathophysiologic factors and ECMO-related complications through central nervous system cellular injury, blood-brain barrier dysfunction (BBB), systemic inflammation and neuroinflammation, and coagulopathy. Plasma biomarkers are an emerging tool used to stratify risk for and diagnose ABI, and prognosticate neurofunctional outcomes.

Background: Three major parties are involved in the hypospadias treatment journey - the patient, their parents/carers, and the surgeon. There is a strong trend towards involving all three, where possible, in deriving evidence around the care pathways. Currently, there are little data available on surgeons' perspectives of distal hypospadias care.

Aims: Hypertrophic cardiomyopathy (HCM) is characterized by unexplained left ventricular hypertrophy (LVH) with key pathologic processes including myocardial necrosis, fibrosis, inflammation, and hypertrophy, which are involved in heart failure (HF), stroke, and even sudden death. Our aim was to explore the communication network among various cells in the heart of transverse aortic constriction (TAC) surgery induced HCM mice.

Materials And Methods: Single-cell RNA-seq data of GSE137167 was downloaded from the Gene Expression Omnibus (GEO) database.
