Isolated yolk platelets, nuclei and pigment granules, ribosomes and the ribosome-free supernatant of homogenates from whole gastrulae ofXenopus laevis were tested for their inducing properties.The subcellular fractions were obtained by differential centrifugation in sucrose media and their purity was checked by electronmicroscopy.The microsomal fraction shows archencephalic and deuterencephalic inducing capacity which is enhanced after extraction with pyrophosphate and with phenol. The membrane structures of this fraction can partially be separated from the ribosomes by treatment with desoxycholate. Proteins with deuterencephalic and spinocaudal inducing activity are extractable from the desoxycholate treated microsomal fraction with 1 M NHCl.The supernatant, free of ribosomes, has a rather weak inducing ability. After extraction with phenol the archencephalic and deuterencephalic activity increases, but not the spinoaudal.Isolated nuclei induce small deuterencephalic structures and a relatively high percentage of spinocaudal differentiations. Extraction with phenol does not enhance the rate of these inductions.The yolk platelets show mainly deuterencephalic inducing activity which is associated essentially with the superficial layer of these particles. Proteins, inducing archencephalic and deuterencephalic tissue differentiations, are extractable in small amounts from the platelets after partial removal of the superficial layer.

We have assessed the type and amount of neural tissue induced in the chick gastrula ectoblast with increasing duration of contact with the Hensen node inducer. At least 4 h contact is necessary to induce the ectoblast in the area pellucida, 9 h in the area opaca and even longer at the margin of overgrowth. In the area pellucida, the inductive response shifts from archencephalic type at 4-6 h contact to deuterencephalic type after 9 h contact.

We have assessed the quality and quantity of the neural inductive response of the chick gastrula ectoblast located at increasing distancefrom the host axis. In a stage 4 gastrula, entire ectoblast exhibits neural competence. The quality of induced neural tissue shifts from deuterencephalic type in the area pellucida to archencephalic type in the area opaca and primitive medullary or palisade type atthe margin of overgrowth with a concomitant reduction in the number of induced neural cells.

Caudalization, which is proposed to be one of two functions of the amphibian organizer, initiates posterior pathways of neural development in the dorsalized ectoderm. In the absence of caudalization, dorsalized ectoderm only expresses the most anterior (archencephalic) differentiation.
