Advances in high-resolution 3D microscopy have enabled the investigation of subcellular microstructures in biological specimen. For a full understanding of the organism's structure and function, it is mandatory to obtain data from the whole animal, not just parts of it. In this paper, we present our work with the Knife-Edge Scanning Microscope (KESM) for imaging a Nissl-stained whole zebrafish larva. KESM combines a diamond microtome and line-scan imaging for simultaneous sectioning and imaging in 3D. We show that using the KESM, a zebrafish, less than 3 mm long and diameter less than 500 μm, can be imaged within 1 hour at a resolution of 0.6 μm × 0.7 μm × 1.0 μm. We also present new results on using a vibrating microtome to improve sectioning and imaging robustness.

We report application of the knife-edge technique at the sharp edges of WS2 and MoS2 monolayer flakes for lateral and axial resolution assessment in all three modalities of nonlinear laser scanning microscopy: two-photon excited fluorescence (TPEF), second- and third-harmonic generation (SHG, THG) imaging. This technique provides a high signal-to-noise ratio, no photobleaching effect and shows good agreement with standard resolution measurement techniques. Furthermore, we assessed both the lateral resolution in TPEF imaging modality and the axial resolution in SHG and THG imaging modality directly via the full-width at half maximum parameter of the corresponding Gaussian distribution.

During operation of an ITER-like plasma fusion device, Plasma Facing Components (PFCs) of a divertor are subjected to steady-state and transient heat loads of up to 20 MW/m2. At High Heat Flux Test Facility (HHFTF) at Institute for Plasma Research, PFCs are subjected to such heat flux scenarios using 200 kW electron beam (EB).
