This study explores the effect of transplantation of undifferentiated Sertoli cells in the testicular tissue of an experimental animal model of bilateral abdominal cryptorchidism. Effectiveness of undifferentiated Sertoli cell transplantation was assessed after 1 and 3 months after injection. Partial restoration of seminiferous tubules was found to occur after transplantation. In a third of them germ cells of all stages of the differentiation were discovered while they were not found in the control group.

For mammalian spermatogenesis to proceed normally, it is essential that the population of testicular progenitor cells, A undifferentiated spermatogonia (A), undergoes differentiation during the A to A1 transition that occurs at the onset of spermatogenesis. The commitment of the A population to differentiation and leaving a quiescent, stem-like state gives rise to all the spermatozoa produced across the lifespan of an individual, and ultimately determines male fertility. The action of all-trans retinoic acid (atRA) on the A population is the determining factor that induces this change.

Mouse spermatogenesis entails the maintenance and self-renewal of spermatogonial stem cells (SSCs), which require a complex web-like signaling network transduced by various cytokines. Although brain-derived neurotrophic factor (BDNF) is expressed in Sertoli cells in the testis, and its receptor tropomyosin receptor kinase B (TrkB) is expressed in the spermatogonial population containing SSCs, potential functions of BDNF for spermatogenesis have not been uncovered. Here, we generate BDNF conditional knockout mice and find that BDNF is dispensable for in vivo spermatogenesis and fertility.

Increasing survival rates of children following cancer treatment have resulted in a significant population of adult survivors with the common side effect of infertility. Additionally, the availability of genetic testing has identified Klinefelter syndrome (classic 47,XXY) as the cause of future male infertility for a significant number of prepubertal patients. This study explores new spermatogonia stem cell (SSC)-based fertility therapies to meet the needs of these patients.
