New streptothricin-group antibiotics, AN-201 I, II and III. II. Chemical structures.

The new, belonging to the streptothricin-group antibiotics AN-201 I, II and III were found to be produced by a soil actinomycete identified as Streptomyces nojiriensis C-13. The chemical structures and the physical and spectroscopic properties of these compounds are reported here. On the basis of NMR and fast atom bombardment mass spectrometry (FAB-MS) spectra the antibiotics were identified as N beta-acetylated derivatives of streptothricins E, D and F.

Four streptothricin-group antibiotics (1~4) were isolated from the fermentation broth of Streptomyces qinlingensis sp. nov. Along with the known antibiotics streptothricins F (1) and D (3), two new members of this class (2, 4) were identified as 12-carbamoyl derivatives of 1 and 3, respectively, mainly by analysis of the IR, HR-MS and NMR spectral data.

Methodical approaches to detecting cultures producing streptothricins at the early stages of screening new antibiotics were developed. The approaches are based on chromatographic and electrophoretic mobility of streptothricins and the products of their hydrolysis in the extracts from agar cultures of actinomycetes. Application of the method for screening new antibiotics is illustrated with an example.
