The innate immune system is the organism's first line of defense against pathogens. Pattern recognition receptors (PRRs) are responsible for sensing the presence of pathogen-associated molecules. The prototypic PRRs, the membrane-bound receptors of the Toll-like receptor (TLR) family, recognize pathogen-associated molecular patterns (PAMPs) and initiate an innate immune response through signaling pathways that depend on the adaptor molecules MyD88 and TRIF. Deciphering the differences in the complex signaling events that lead to pathogen recognition and initiation of the correct response remains challenging. Here we report the discovery of temporal changes in the protein signaling components involved in innate immunity. Using an integrated strategy combining unbiased proteomics, transcriptomics and macrophage stimulations with three different PAMPs, we identified differences in signaling between individual TLRs and revealed specifics of pathway regulation at the protein level.

The aggregation state of nano-TiO in the environment is altered under marine heatwaves (MHWs), thus affecting its bioavailability and toxicity to the marine organisms. Here, we investigated the toxic mechanisms and effects of nano-TiO on gut-hepatopancreas axis health of Mytilus coruscus exposed to 25 and 250 μg/L of nano-TiO under laboratory-simulated MHW. Compared with the control conditions or post-MHW cooling phase, prolonged MHW exposure significantly inhibited digestive function, decreased immune-related enzymes activities, and caused neurotoxicity in the mussels.

Cytokine storm is a life-threatening systemic hyper-inflammatory state caused by different etiologies, in which the bulk production of pro-inflammatory cytokines from activated macrophages has a central role. Integrated stress response (ISR) comprises several protective signaling pathways, leading to phosphorylation of eukaryotic initiation factor 2α (eIF2α) and repression of protein translation. Emerging evidence suggests that ISR induction may elicit anti-inflammatory effects.
