Isomerization of aspartic acid residues is a relevant degradation pathway of protein biopharmaceuticals as it can impair their biological activity. However, the in silico prediction of isomerization hotspots and their consequences remains ambiguous and misleading. We have previously shown that all ion differential analysis (AiDA) of middle-down spectra can be used to reveal diagnostic terminal and internal fragments with more sensitivity than the conventional fragment ion mass matching methodology.

Comparative glycomics data are compositional data, where measured glycans are parts of a whole, indicated by relative abundances. Applying traditional statistical analyses to these data often results in misleading conclusions, such as spurious "decreases" of glycans when other structures increase in abundance, or high false-positive rates for differential abundance. Our work introduces a compositional data analysis framework, tailored to comparative glycomics, to account for these data dependencies.

This work represents a significant contribution to understanding the importance of appropriately rounding numbers with minimal error. That is, to reduce inexact rounding and data truncation error and simultaneously eliminate unintentional misleading findings in epidemiological studies. The rounding of numbers represents a compromise solution that attempts to find a balance between the loss of information from reporting too few significant digits versus retaining more digits than necessary.
