A dual mode fluorescent probe, which is based on an integration of fluorescein and coumarin fluorophores, was developed for the discrimination of Cys from Hcy and GSH. This probe (2) shows the advantage of quick reaction (5 min) with Cys, resulting in a strong fluorescence turn-on response when excited at 450 nm. Notably, it also demonstrates the ratiometric fluorescence property while excited by a shorter wavelength (332 nm). All of results suggest probe 2 has a high selectivity toward Cys even in the presence of other amino acids, cations and anions. The detection limit of Cys was calculated as 0.084 μM, which was much lower than the intracellular concentration. H NMR, MS and DFT calculation were used to reveal the detection mechanism further. Finally, this low cytotoxic probe was successfully applied in bioimaging within HepG2 cells.

A new Rhodamine functionalised Schiff Base sensor 3',6'-bis(diethylamino)-2-((4-hydroxybenzylidene)amino)spiro[isoindoline-1,9'-xanthen]-3-one (SBRB1) was designed and synthesized. The recognition ability of sensor SBRB1 towards Hg was studied by using UV-Vis and fluorescence spectroscopy. The fluorescence results showed that the sensor SBRB1 has specific selectivity as well as sensitivity towards Hg among other competitive metal ions as the fluorescence intensity at 479 nm quenched only in the presence of Hg.

Ultrasmall and highly fluorescent gold nanoclusters (Au NCs) have been widely used for the construction of sensing and imaging platforms. Specifically, through a combination of surface functionalization and spectral analysis and/or imaging techniques, effective intracellular detection and imaging are realized. In this review, we summarize the recently adopted intracellular analysis and imaging events with Au NCs-based probes.

Background: Based on the low volatility and refractory nature of Tetracycline (TC), excessive use leads to its continuous accumulation in water environments, posing serious risks to the ecological environment and human health. Although a very limited number of nanomaterials capable of simultaneously detecting and removing TC have been fabricated, they generally exist issues associated with a single detection signal ("on" or "off") or low adsorption rates with low adsorption capacities. As a result, it is crucial to develop a reliable technique to achieve ratiometric detection as well as rapid and efficient removal of TC.

A ratiometric fluorescent nanoprobe (CDs-Rho), synthesized through the simple covalent amide linkage between carbon dots (CDs) and pH-sensitive rhodamine dye (Rho), was designed for the precise sensing and imaging of extremely alkaline environments. The sensing mechanism involves the opposite pH-dependent fluorescence changes in CDs and Rho, respectively, coupled with pH-regulated FRET efficiency from CDs to Rho. The nanoprobe features a wide pH response window from pH 7.

As one of the essential components of reactive oxygen species (ROS), peroxynitrite (ONOO-) plays an indispensable role in redox homeostasis and signal transduction processes, and its deviant levels are associated with numerous clinical diseases. Therefore, accurate and rapid detection of intracellular ONOO- levels is crucial for revealing its role in physiological and pathological processes. Herein, we constructed a ratiometric fluorescent probe to detect ONOO- levels in biological systems.
