By setting the regulatory-approved protocol for a suite of first-in-human studies on BIA 10-2474 against the subsequent French investigations, we highlight 6 key design and statistical issues, which reinforce recommendations by a Royal Statistical Society Working Party, which were made in the aftermath of cytokine release storm in 6 healthy volunteers in the United Kingdom in 2006. The 6 issues are dose determination, availability of pharmacokinetic results, dosing interval, stopping rules, appraisal by safety committee, and clear algorithm required if combining approvals for single and multiple ascending dose studies.

Understanding the biomechanics of osteoarthritis is necessary for designing a biomedical knee implant to reduce pain, increase mobility, and enhance the patient's quality of life. The most appropriate implant design may be chosen by using Multi-Attribute Group Decision-Making (MAGDM) techniques, which include a number of variables including material characteristics, biomechanical performance, cost-effectiveness, and patient-specific requirements. Compared to conventional fuzzy set structures, Spherical Fuzzy -Number Sets ( S) provide an enhanced method for resolving uncertainty in MAGDM and are more suited for handling complicated decision-making situations.

Background: Only 15% of the nearly 30 million Americans with hearing loss use hearing aids, partly due to high cost, stigma, and limited access to professional hearing care. Hearing impairment in adults can lead to social isolation and depression and is associated with an increased risk of falls. Given the persistent barriers to hearing aid use, the Food and Drug Administration issued a final rule to allow over-the-counter hearing aids to be sold directly to adult consumers with perceived mild to moderate hearing loss at pharmacies, stores, and online retailers without seeing a physician or licensed hearing health care professional.
