Problem: We investigated the effect of palmitic acid (PA), a major saturated fatty acid in NEFA, on bovine oviduct epithelial cells (OECs) during in vitro cell culture.

Method Of Study: Bovine oviductal tissues ipsilateral to the corpus luteum were collected 1-3 days after ovulation; the OECs were isolated and cultured.

Results: PA increased lipid accumulation and activated caspase-3 in OECs, resulting in decreased cell proliferation. PA also stimulated the secretion of inflammatory cytokine interleukin (IL)-8 depending on TLR4, NF-κB activation, and reactive oxygen species (ROS) production. Moreover, PA induced mitochondrial dysfunction, including mitochondrial fission, ATP production, and mitochondrial ROS production. It also increased levels of LC3 and p62 proteins, suggesting autophagy induction in OECs.

Conclusion: We suggest that bovine OECs recognize an excessive increase in endogenous and sterile danger signals, such as PA, which may contribute to chronic oviductal inflammation, resulting in infertility associated with oviductal dysfunction.

Background: Knowledge about the diet quality among youth who follow different types of plant-based diets is essential to understand whether support is required to ensure a well-planned diet that meets their nutritional needs. This study aimed to investigate how food groups, macronutrient intake, and objective blood measures varied between Norwegian youth following different plant-based diets compared to omnivorous diet.

Methods: Cross-sectional design, with healthy 16-to-24-year-olds (n = 165) recruited from the Agder area in Norway, following a vegan, lacto-ovo-vegetarian, pescatarian, flexitarian or omnivore diet.

Objectives: To investigate the regulatory role of nucleotide-bound oligomerized domain-like receptor containing pyrin-domain protein 6 (NLRP6) in liver lipid metabolism and non-alcoholic fatty liver disease (NAFLD).

Methods: Mouse models with high-fat diet (HFD) feeding for 16 weeks (=6) or with methionine choline-deficient diet (MCD) feeding for 8 weeks (=6) were examined for the development of NAFLD using HE and oil red O staining, and hepatic expressions of NLRP6 were detected with RT-qPCR, Western blotting, and immunohistochemical staining. Cultured human hepatocytes (LO2 cells) with adenovirus-mediated NLRP6 overexpression or knock-down were treated with palmitic acid (PA) in the presence or absence of compound C (an AMPK inhibitor), and the changes in cellular lipid metabolism were examined by measuring triglyceride, ATP and β-hydroxybutyrate levels and using oil red staining, RT-qPCR, and Western blotting.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a prevalent hepatic disorder worldwide. Arachidonic acid 15-lipoxygenase (ALOX15), an enzyme catalyzing the peroxidation of polyunsaturated fatty acids, plays a crucial role in various diseases. Here, we sought to investigate the involvement of ALOX15 in MASLD.

In clinical practice, thymopentin (TP-5) is a commonly utilized immunomodulatory peptide drug. The relatively short half-life of TP-5, however, significantly limits its applicability in immunotherapy. Inspired by the structure of the TLR2 ligand lipopeptide Pam3CSK4, fatty acid-modified TP-5 peptides were designed and synthesized in this study.

Background: The liver plays a central role in fat storage, but little is known about physiological fat accumulation during early development. Here we investigated a transient surge in hepatic lipid droplets observed in newborn mice immediately after birth.

Methods: We developed a novel model to quantify liver fat content without tissue processing.
