Activity and selectivity assessment of new bi-aryl amide 11β-hydroxysteroid dehydrogenase 1 (11β-HSD1) inhibitors, prepared in a modular manner via Suzuki cross-coupling, are described. Several compounds inhibiting 11β-HSD1 at nanomolar concentrations were identified. Compounds 2b, 3e, 7b and 12e were shown to selectively inhibit 11β-HSD1 over 11β-HSD2, 17β-HSD1 and 17β-HSD2. These inhibitors also potently inhibited 11β-HSD1 activity in intact HEK-293 cells expressing the recombinant enzyme and in intact primary human keratinocytes expressing endogenous 11β-HSD1. Moreover, compounds 2b, 3e and 12e were tested for their activity in human skin biopsies. They were able to prevent, at least in part, both the cortisone- and the UV-mediated decreases in collagen content. Thus, inhibition of 11β-HSD1 by these compounds can be further investigated to delay or prevent UV-mediated skin damage and skin aging.

A novel analytical method was designed and developed that exhibited ultraviolet-visible (UV-Vis), fluorescence (FL), and resonance Rayleigh scattering (RRS) signals for straightforward and comprehensive determination of monoamine oxidase B (MAO-B) using polyethylenimine-functionalized silver nanoparticles (PEI-Ag NPs). Through a facile one-step experiment, and NaOH assisted, in an aqueous solution of 100 ℃ for 40 min PEI reacted with AgNO to generate PEI-Ag NPs with a yellow color and weak blue fluorescence. Interestingly, phenylacetaldehyde (PAA), a specific product of MAO-B, causes significant enhancement of the three optical signals of UV-Vis, FL, and RRS.

A new strategy has been developed to successfully produce the active component danshensu ex vivo. For this purpose, phenylalanine dehydrogenase from Bacillus sphaericus was combined with the novel hydroxyphenylpyruvate reductase from Mentha x piperita, thereby providing an in situ cofactor regeneration throughout the conversion process. The purified enzymes were co-immobilized and subsequently employed in batch biotransformation, resulting in 60% conversion of 10 mM L-dopa within 24 h, with a catalytic amount of NAD as cofactor.
