Larsen syndrome (LRS) is a rare genetic disease associated with variable manifestations including skeletal malformations, dislocations of the large joints, and notable changes in facial and limb features. Genetic variants in the Filamin B (FLNB) gene are associated with the development of LRS. We searched two literature databases (OMIM and PubMed) and three gene variant databases (HGMD, UniProt, & dbSNP) to capture all the possible variants associated with LRS phenotype, which may have an impact on the FLNB function. Our search yielded 77 variants that might impact the FLNB protein function in patients with LRS. We performed rigorous computational analysis such as conservational, biochemical, pathogenicity, and structural computational analyses to understand the deleterious effect of the G1691S variant. Further, the structural changes of the G1691S variant was compared with a null variant (G1691A) and the native protein through a molecular dynamic simulation study of 50 ns. We found that the variant G1691S was highly deleterious and destabilize the protein when compared to the native and variant G1691A. This might be due to the physicochemical changes in the variant G1691S when compared to the native and variant G1691A. The destabilization was further supported by transformation of bend to coil in variant G1691S whereas bend was retained in native and variant G1691A through molecular dynamics analysis. Our study shed light on the importance of computational methods to understand the molecular nature of genetic variants and structural insights on the function of the FLNB protein. J. Cell. Assessment of Genetic Variants Linked to Susceptibility to Mechanical Prosthetic Valve Thrombosis.

Am J Cardiol

January 2025

Department of Cardiology, Koşuyolu Kartal Heart Training and Research Hospital, Istanbul, Türkiye; Division of Health Sciences, Ardahan University, Ardahan, Türkiye.

Prosthetic valve thrombosis (PVT) is a critical and life-threatening condition driven by multifactorial etiologies, including genetic predispositions. The study was designed as a single-center retrospective manner. Echocardiographic features and genetic test including factor II/prothrombin (G20210A), factor V Leiden (G1691A), factor V R2 (A4070G), apolipoprotein (Apo) B-100 (G10708A), ApoE (C112R), ApoE (R158C), methylenetetrahydrofolate reductase (MTHFR) C677T, MTHFR A1298C, factor XIII G103T (V34L), β-fibrinogen (455G>A), PAI-1 4G/5G, and HPA-1 GPIIIa (T196C) genotyping variations were assessed.

Background/aim: We aimed to determine the genetic risk factors in patients aged 45 years and below with a history of early myocardial infarction (MI), compared to individuals over 60 years of age with no history of MI.

Materials And Methods: In this study, we selected different age groups to more clearly distinguish genetic differences. Accordingly, we compared individuals who had experienced MI at an early age with those who were older and had not experienced any cardiovascular events.

Article Synopsis
  • - This study examined how genetic thrombophilia affects early versus late pregnancy loss in 156 women, dividing them into two groups based on when they experienced miscarriage.
  • - Key genetic mutations were identified as risk factors for pregnancy loss, with certain combinations, like Factor V Leiden and MTHFR mutations, showing significant interactions linked to early loss.
  • - Various mutations had different odds ratios indicating their risk levels, with some being more associated with early pregnancy loss (like PAI-1 4G/5G) and others (like MTHFR A1298C) linked to late pregnancy loss.

Article Synopsis
  • A study investigated the link between specific genetic polymorphisms and ST elevation Myocardial Infarction in young Mexican individuals, involving 350 patients under 45 and 350 matched controls.
  • The A1166C polymorphism was found to significantly increase the risk of Myocardial Infarction, while G20210A, G1691A, 97G > T, and A1298C did not show a similar association.
  • Other factors like dyslipidemia, hypertension, smoking, and family history were also linked to increased risks, indicating that genetic variations might contribute to early cardiovascular issues, but more research is needed on gene interactions.

Background: In recent years, many studies have found possible links between gene polymorphisms and venous thromboembolism (VTE). By identifying genetic risk factors before facing environmental risk factors such as surgical interventions and COVID-19 vaccination, we could rapidly respond to the risk of VTE. The aim of this study was to perform an umbrella review of genetic variants related to VTE.
