Characterization of glial-restricted precursors from rhesus monkey embryonic stem cells.

Glial-restricted precursor (GRP) cells, the earliest glial progenitors for both astrocytes and oligodendrocytes, have been derived from embryos and embryonic stem cells (ESC) in rodents. However, knowledge regarding the equivalent cell type in primates is limited due to restrictions imposed by ethics and resources. Here we report successful derivation and characterization of primate GRP cells from rhesus monkey ESC. The purified monkey GRP cells were AB-positive and FGF2-dependent for survival and proliferation. The differentiation assays indicated that they were tri-potential and bi-potential . These newly purified GRP cells will help to facilitate understanding of the molecular mechanism of glial development in primates as well as provide a source of therapeutic donor cells for use in neuroregenerative medicine.

Background: Whereas several studies have demonstrated the long-term immunogenicity of BNT162b2 vaccine in healthy adults, little evidence was provided in vulnerable populations (VPs) with generally low ability to respond to immunizations. We aimed to identify pre-vaccination immune phenotype and transcriptional signatures in B and T-cell subsets associated with immune response and long-term maintenance upon SARS-CoV-2 vaccination in VPs.

Methods: A cohort of VPs (N = 169) including solid organ transplant recipients (SOT; N = 35), Inflammatory Bowel Disease (N = 31), Down Syndrome (N = 42), people living with HIV (N = 36), primary immune deficiencies (N = 25) and healthy controls (N = 37) were enrolled in the CONVERS Study.

To investigate the heterogeneity of fibroblasts in human fetal skin, we analyzed published single-cell RNA-sequencing data (7 and 16 weeks after conception) and performed single-molecule FISH to map their spatial distribution and predicted dynamic interactions. Clustering revealed 8 fibroblast populations with changes in developmental stage-specific abundance. Proliferative cells (MKI67+) were present at all stages.
