Carboxylesterases (CEs) are ubiquitous enzymes that are responsible for the metabolism of xenobiotics, including drugs such as irinotecan and oseltamivir. Inhibition of CEs significantly modulates the efficacy of such agents. We report here that β-lapachone is a potent, reversible CE inhibitor with K values in the nanomolar range. A series of amino and phenoxy analogues have been synthesized, and although the former are very poor inhibitors, the latter compounds are highly effective in modulating CE activity. Our data demonstrate that tautomerism of the amino derivatives to the imino forms likely accounts for their loss in biological activity. A series of N-methylated amino derivatives, which are unable to undergo such tautomerism, were equal in potency to the phenoxy analogues and demonstrated selectivity for the liver enzyme hCE1. These specific inhibitors, which are active in cell culture models, will be exceptionally useful reagents for reaction profiling of esterified drugs in complex biological samples.

Introduction: Phenolic compounds garner interest in developing medicines, nutraceuticals, and cosmeceuticals based on natural products. The quantity of phenolic compounds in a sample is commonly determined via spectrophotometry; however, this instrumented technique is relatively laborious and time consuming and requires a large amount of reagents.

Objective: This work aimed to develop a simple, point-of-need colorimetric sensor to rapidly determine total phenolic content (TPC) in tea extracts.

Polymerase chain reactions (PCR) are most reliable and precise means for nucleic acid analysis of biological samples. A cold-chain system with temperature at around -20°C is generally necessary for storage and transportation of PCR-related reagents. In order to facilitate ambient temperature storage and transportation, this study prepared Taq DNA polymerase and 5 × HS-Taq Mix (as low as 0.

To alleviate the energy crisis and control environmental pollution raised by spent lithium-ion batteries (LIBs), the development of efficient and economic methods for their recycling is crucial for sustainable development of new energy industry. Herein, a combined pyro - hydrometallurgical process was adopted for recovery of valuable metal elements for spent LiNiCoMnO (NCM523). Different from conventional pyrometallurgical methods with high temperature and energy consumption, the NHHSO roasting strategy works at 400 °C and achieves remarkable leaching efficiencies of Li, Co, Mn, and Ni achieved 97.

Silver nanowire/gold nanosphere binary plasma-assembled membranes for sensitive SERS detection of homocysteine.

Mikrochim Acta

December 2024

School of Materials and Chemical Engineering, Fuzhou Institute of Oceanography, Minjiang University, Fuzhou, Fujian, 350108, China.

Silver nanowire (Ag NW)/gold nanosphere (Au NS) binary plasma films were prepared using plasma coupling between Ag NWs and Au NSs. The plasma films formed by combining these two noble metals showed better sensitivity for SERS detection with a minimum detection concentration of 10 M for R6G compared to pure Ag NWs or Au NSs. After rational optimisation of the substrate preparation process, the substrate showed good homogeneity, reproducibility and stability.
