The present study investigated whether the presence or absence of peripubertal ovarian hormones affects sexual preference and conditioned place preference for paced mating in adult female rats primed with 10μg estradiol benzoate and 1mg progesterone. Ovariectomy (OVX) occurred either before or after pubertal development, and 4weeks later rats began a series of behavioral tests. Rats with ovaries removed before the pubertal timeframe (Prepubertal OVX) were more active, more likely to withdrawal from the male compartment, and did not discriminate between mounts and intromissions during paced mating relative to rats with ovaries during puberty (Adult OVX). Both Adult OVX and Prepubertal OVX rats showed a higher preference for the male when hormone primed vs. oil treated and a conditioned place preference for paced mating behavior. The results of the present study demonstrate that some, but not all, aspects of female sexual behavior require ovarian hormones during puberty.

Kisspeptin (Kp), a potent regulator of the hypothalamic-pituitary-gonad axis, was recently shown to be involved in partner preference and sexual receptivity in females. Interestingly, Kp and its receptor (Kiss1r) are expressed in brain regions involved in the reward and motivation of reinforcing behaviors. Therefore, in the present study, we designed 3 experiments to determine the participation of Kp in female sexual behavior and the positive affective (PA) reward state induced by paced mating (PM).

Sexual behavior in female rats varies depending on sexual history and the combination of ovarian hormones administered to induce receptivity. Experiment 1 tested whether paced mating behavior differed in sexually experienced rats when receptivity was induced with sequential estradiol benzoate (EB) and progesterone (P) or EB-Alone. Rats gained paced mating experience under EB/P (10 μg EB 48 hr + 1 mg P 4-6 hr before mating) and then were primed with EB-Alone (2 μg EB for 6 days).

The relationship between diabetes mellitus type 2 (DM2) and sexual desire in women has not been systematically studied, therefore, animal models have been used for this purpose. When streptozotocin (STZ) is administered in the neonatal stage, the rat shows moderate chronic hyperglycemia and glucose intolerance in adulthood, resembling a DM2 model. These females show less alterations of sexual behavior (a slight decreased proceptivity and loss of paced mating) than their counterpart with severe hyperglycemia.
