Determining sensor parameters is a prerequisite for quantitative force measurement. Here we report a direct, high-precision calibration method for quartz tuning fork (TF) sensors that are popular in the field of nanomechanical measurement. In the method, conservative and dissipative forces with controlled amplitudes are applied to one prong of TF directly to mimic the tip-sample interaction, and the responses of the sensor are measured at the same time to extract sensor parameters. The method, for the first time, allows force gradient and damping coefficient which correspond to the conservative and dissipative interactions to be measured simultaneously. The calibration result shows surprisingly that, unlike cantilevers, the frequency shift for TFs depends on both the conservative and dissipative forces, which may be ascribed to the complex dynamics. The effectiveness of the method is testified by force spectrum measurement with a calibrated TF. The method is generic for all kinds of sensors used for non-contact atomic force microscopy (NC-AFM) and is an important improvement for quantitative nanomechanical measurement.

Nucleoside triphosphate (NTP)-dependent protein assemblies such as microtubules and actin filaments have inspired the development of diverse chemically fueled molecular machines and active materials but their functional sophistication has yet to be matched by design. Given this challenge, we asked whether it is possible to transform a natural adenosine 5'-triphosphate (ATP)-dependent enzyme into a dissipative self-assembling system, thereby altering the structural and functional mode in which chemical energy is used. Here we report that FtsH (filamentous temperature-sensitive protease H), a hexameric ATPase involved in membrane protein degradation, can be readily engineered to form one-dimensional helical nanotubes.

As opportunistic generalists occupying a range of ecological niches, chacma baboons (Papio ursinus) are considered a highly flexible species of relatively low conservation priority. Underlying their ecological flexibility is a repertoire of behavioral strategies observed in response to ecological stressors. Although these strategies are relatively well-documented, we know very little about how they impact upon an individual's thermal and energetic physiology, which can influence population-level reproductive potential in the face of climatic warming.

Role of Abscisic Acid in the Whole-Body Regulation of Glucose Uptake and Metabolism.

Abscisic acid (ABA) is a hormone with a long evolutionary history, dating back to the earliest living organisms, of which modern (ABA-producing) cyanobacteria are likely descendants, which existed long before the separation of the plant and animal kingdoms, with a conserved role as signals regulating cell responses to environmental challenges. In mammals, along with the anti-inflammatory and neuroprotective function of ABA, nanomolar ABA regulates the metabolic response to glucose availability by stimulating glucose uptake in skeletal muscle and adipose tissue via an insulin-independent mechanism and increasing metabolic energy production and also dissipation in brown and white adipocytes. Chronic ABA intake of micrograms per Kg body weight improves blood glucose, lipids, and morphometric parameters (waist circumference and body mass index) in borderline subjects for prediabetes and metabolic syndrome.
