Evaluation of TOCSY mixing for sensitivity-enhancement in solid-state NMR and application of 4D experiments for side-chain assignments of the full-length 30 kDa membrane protein GlpG.

J Biomol NMR

January 2025

Research Unit Molecular Biophysics, Leibniz-Forschungsinstitut für Molekulare Pharmakologie, Robert- Rössle-Straße 10, 13125, Berlin, Germany.

Chemical shift assignments of large membrane proteins by solid-state NMR experiments are challenging. Recent advancements in sensitivity-enhanced pulse sequences, have made it feasible to acquire H-detected 4D spectra of these challenging protein samples within reasonable timeframes. However, obtaining unambiguous assignments remains difficult without access to side-chain chemical shifts.

Autologous platelet concentrates as adjuncts to non-surgical periodontal therapy: a systematic review and meta-analysis.

Clin Oral Investig

January 2025

Periodontology Unit, Centre for Host Microbiome Interactions, Faculty of Dentistry, Oral & Craniofacial Sciences, King's College London, London, UK.

Objective: To evaluate the possible additional clinical benefit from autologous platelet concentrate (APC) treatment adjunct to non-surgical periodontal therapy (NSPT).

Methods: Electronic (MEDLINE/Embase/Cochrane/MedNar/CORE) and hand searches were conducted. Following studies selection, evidence tables were formed, and meta-analyses were performed for the following outcomes: probing pocket depth (PPD) reduction, clinical attachment level (CAL) gain, and bleeding on probing (BoP) reduction.

Polyelectrolyte brushes (PEBs) undergo conformational transitions due to changes in pH and/or ionic strength, which is leveraged as smart surfaces and on-demand drug-release systems. However, probing conformational transitions of functional PEBs has remained challenging due to low spatiotemporal resolution of characterization methods. Herein, fluorescently-coupled PEBs are devised that give rise to Förster Resonance Energy Transfer (FRET) intrinsically coupled to conformational transitions of chains.

TRAF2 and RIPK1 redundantly mediate classical NFκB signaling by TNFR1 and CD95-type death receptors.

Cell Death Dis

January 2025

Division of Molecular Internal Medicine, Department of Internal Medicine II, University Hospital Würzburg, Auvera Haus Grombühlstraße 12, 97080, Würzburg, Germany.

This study suggests a modified model of TNFR1-induced complex I-mediated NFκB signaling. Evaluation of a panel of five tumor cell lines (HCT116-PIK3CAmut, SK-MEL-23, HeLa-RIPK3, HT29, D10) with TRAF2 knockout revealed in two cell lines (HT29, HeLa-RIPK3) a sensitizing effect for death receptor-induced necroptosis and in one cell line (D10) a mild sensitization for TNFR1-induced apoptosis. TRAF2 deficiency inhibited death receptor-induced classical NFκB-mediated production of IL-8 only in a subset of cell lines and only partly.

Effects of laser wavelength and pulse energy on the evaporation behavior of TiN coatings in atom probe tomography: A multi-instrument study.

Ultramicroscopy

January 2025

Christian Doppler Laboratory for Sustainable Hard Coatings at the Department of Materials Science, Montanuniversität Leoben, Franz-Josef-Straße 18, 8700 Leoben, Austria.

The impact of the laser wavelength on accuracy in elemental composition analysis in atom probe tomography (APT) was investigated. Three different commercial atom probe systems - LEAP 3000X HR, LEAP 5000 XR, and LEAP 6000 XR - were systematically compared for a TiN model coating studying the effect of shorter laser wavelengths, especially in the deep ultraviolet (DUV) range, on the evaporation behavior. The findings demonstrate that the use of shorter wavelengths enhances the accuracy in elemental composition, while maintaining similar electric field strengths.
