For accurate micro-scale quantification of a specific protein in biological fluids, immunoaffinity chromatography (IAC) and isoelectric focusing (IEF) were combined in a single fused-silica capillary. The inner wall of the capillary was coated with an anti-E-tag antibody at the inlet side to form an IAC column, and polydimethylacrylamide, a neutral polymer, at the outlet side to form the capillary for IEF. After loading a sample, the whole capillary was filled with a carrier ampholyte solution. An anode solution, an acid, was then introduced to fill only the IAC column segment. Focusing was started with a pressure that balances with the electroosmotic flow produced in the acidified IAC column. Fluorescence-labeled recombinant Fab with an E-tag spiked at 16 pM to 10 nM in 50% serum was separated and detected with high precision. The coupling principle allows rapid and high-resolution IEF analysis of a protein in a biological sample without any loss of the immunoaffinity captured protein.

Background: The 11+Myco MS-PREP® Immunoaffinity Column (IAC) contains a gel suspension of monoclonal antibodies specific to the toxins of interest. Following sample extraction, the IAC is used for cleanup and concentration of mycotoxins prior to analysis by Liquid Chromatography with Tandem Mass Spectrometry (LC-MS/MS).
Objective: This study evaluated the IAC with LC-MS/MS method for Performance Tested Methods SM certification for simultaneous determination and confirmation of Aflatoxins B1, B2, G1, G2, and M1; Deoxynivalenol, Fumonisins B1, B2, and B3; Ochratoxin A; T-2; HT-2; and Zearalenone from corn, wheat, cereal-based baby food (with and without dairy ingredients), paprika, chili powder and animal feed.

Silk was the driving force behind the opening of the Silk Road, positioning this material as a global commodity that greatly influenced the progress of human civilization. Due to the natural protein properties of silk, the internal structure and external characteristics of silk cultural relics are irreversibly destroyed during the process of burial or when passed down through the generations until the production information and material components completely disappear, making it difficult to obtain direct archaeological evidence for pinpointing the origin of silk. The sacrificial pits at the Sanxingdui archaeologcal site, located in Guanghan, Sichuan, China, have been found with layers of ash above the layers of artifacts and some bronzes with fabric traces.
